Euphorbia fusiformis Buch.-Ham.Ex D.Don, (Synonym: Euphorbia acaulis Roxb) is an infrequent medicinal, geophytic herb belonging to family Euphorbiaceae. The latex obtained from tubers of Euphorbia fusiformis was traditionally used as an antidote in scorpion and snake bites. Fresh latex has been externally applied to heal chronic wounds and cracks and to cure skin diseases. Previous research on the Euphorbia fusifornis primarily focused on phytochemical investigation and pharmacological screening of various solvent extracts of dried root powder. Considering the therapeutic potential of the latex, there is a need to explore the studies on plant latex. The present work deals with the determination of physicochemical properties, preliminary phytochemical analysis and acute oral toxicity study of latex of Euphorbia fusiformis. Physicochemical characterization and preliminary phytochemical studies were carried out by using ethanolic solution of latex. The studies revealed the presence of alkaloids, glycosides, terpenoids, steroids and saponins. Acute oral toxicity study of latex were carried out by acute toxic class method of OECD 423 guideline. Euphorbia fusiformis latex was found to be safe at the dose of 2000 mg/kg and therefore characterized as Category 5 in the GHS system. It is concluded that further studies are essential for isolation and identification of chemical constituents from Euphorbia fusiformis and screening of pharmacological activities to support the traditional claims. © RJPT All right reserved.

Preliminary Qualitative Phytochemical Analysis and Acute Oral Toxicity Study of Latex of an Ethnomedicinal Plant Euphorbia fusiformis Buch.-Ham.Ex D.Don

This research is focused on designing, synthesis and biological evaluation of a series of coumarin based benzothiazole derivatives. The ligands were identified by docking study for antioxidant and antibacterial potential using target proteins PDB:4H1J and PDB:3G75, respectively. The target molecules were synthesized as a series of substituted N-(benzothiazol-2-yl)-2-oxo-chromene-3-carboxamides (7a–h) by condensation of substituted benzo[d]thiazol-2-amines with in situ synthesized substituted 2-oxo-2H-chromene-3-carbonyl chlorides. Infrared spectroscopy and 1H- nuclear magnetic resonance spectra were used to characterize the synthesized molecules. In vitro antioxidant activity of compounds was evaluated by DPPH and H2O2 radical scavenging assays. Antibacterial potential of compounds was evaluated using well diffusion method against Staphylococcus aureus (ATCC 25923), Escherichia coli (ATCC 25922) and Pseudomonas aeruginosa (ATCC 27853). Among synthesized derivatives, 7a showed good antioxidant potential whereas 7f showed antibacterial activity, which might be employed as lead molecules for future investigation for respective activities. © 2022 Indian Drug Manufacturers' Association. DESIGN, SYNTHESIS AND EVALUATION OF N-(BENZO[D]THIAZOL-2-YL)-2-OXO-2H-CHROMENE-3-CARBOXAMIDE DERIVATIVES AS POTENTIAL ANTIOXIDANT AND ANTIBACTERIAL AGENTS

The selective optimization of side activities (SOSA) approach appears to be a promising strategy for lead generation. In this approach old drugs are used to generate new hits or leads. The objective of SOSA is to prepare analogues of the hit molecule in order to transform the observed ``side activity{''} into the main effect and to strongly reduce or abolish the initial pharmacological activity. The idea of taking a molecule with a primary activity in humans and then enhancing a secondary effect through structural changes describes the most common implementation of SOSA. An advantage to starting a drug discovery program with molecules that have already been tested in humans is that those molecules have already satisfied many safety criteria. Such molecules also likely have favourable pharmacokinetic profiles. In the present review different successful examples of SOSA switches are summarized. We hope that the present review will be useful for scientists working in the area of drug design and discovery.

Selective Optimization of Side Activities (SOSA) as an Efficient Approach for Generation of New Leads from Old Drugs

Post injury, healing of wound is essential for recovery of uprightness of the body, which is one of the complex, continuous and unanticipated chains of events in case of diabetic patients. Nitric oxide represents a potential wound therapeutic agent due to its ability to regulate inflammation and eradicate bacterial infections. Impaired wound healing is a prominent diabetic complication which may lead to amputations also. In addition to modern medicines we can use nitric oxide therapy prominently for diabetic wound healing. Prominent and proven role of nitric oxide as well as conventional materials (like metformin and hydrogen sulphide, whey proteins, acidified nitrile etc), therapies (like low level laser therapy, hyperbaric oxygen therapy etc) and techniques (like in vivo implants with biosensors) can be taken into consideration. Many plant extracts showed promising results for wound healing activity by increasing nitric oxide levels. Use of modern technologies such as implant with biosensor and technique like sonic head hog gene are available for diabetic wound healing using Nitric oxide. In this review, an attempt has been made to compile comprehensive updated information of role of nitric oxide in diabetic wound healing, which may be exploited by focusing more on development of effective strategies to treat diabetes-associated wound.
