Remediation of Medication Error : Salience of Labelling

Intranasal drug administration provides a practical and safe way of reaching the brain parenchyma, bypassing the blood-brain barrier (BBB), within minutes. It is also an excellent strategy to deliver the drugs directly to the brain, eliminating the necessity for systemic administration, thereby reducing the potential side effects of neuroactive therapeutics. The unique connections between the brain and external environment, mediated through olfactory and trigeminal nerves, allow the direct naso-brain delivery of potent neurotherapeutics. This targeted drug delivery system has a huge potential for commercialization, as it allows an easy attainment of the effective drug dose at the target site. This chapter will be an important bunch of literature illustrating the major barriers to an effective naso-brain delivery, and the key physicochemical, biopharmaceutical, and practical considerations need attention by the researchers while formulating the efficient nose-to-brain drug delivery. © 2021 Elsevier Inc.

A Mini Review - Pharmaceutical Creams

The sustained release matrix dosage type is favored to prevent blood level variations found in the Dipyridamole drug. The goal of this research was to formulate Dipyridamole's sustained release matrix tablet, Dipyridamole is a thromboembolic risk preventive drug for heart valve replacement and long-term angina pectoris treatment will be well absorbed in the stomach. Maintaining long-term therapeutic plasma concentration and increasing bioavailability by using various polymers to improve bioavailability and to minimize the dosing rate and side effects by integrating a 3-factor, 3-level Box-Behnken statistical design of surface response technique. Sustain release polymers such as HPMC K4M (X1), MCC 101 (X2) and Magnesium Stearate (X3) are the dependent variables and Independent variable is the percentage release of drug at 12 h (Y1) were determined. Response surface plots of Box-Behnken design have been drawn, quadratic models have been statistically validated and optimized formulations have been selected Based on grid search and feasibility. The physical evaluation, drug content and % drug release tests were conducted on all the 13 box Behnken design runs. Zero order, first order, Higuchi and Korsemayer-peppas in terms of r2 and n-value were generated using various release kinetic models and equations. The response surface plots showed high degree of prediction. The desired batch depicted a steady and sustained release from the confirmatory runs (best fit model– Higuchi model (n=0.9862). Hence, the bioavailability of Dipyridamole cocrystal sustained release matrix tablets was increased using response surface designs.

Development and evaluation of dipyridamole matrix tablets using response surface methodology

We have designed and synthesized (Z)-2-(5-methyl-2-oxo-2H-chromen-7-yl) oxy)-N'-(2-oxoindolin-3-ylidin) acetohydrazide derivatives by reacting 7-hydroxy-4-methyl-coumarin and substituted isatin to afford 12 substituted coumarin acetohydrazide derivatives. The synthesized compounds of coumarin acetohydrazide derivatives were designed and evaluated to study for their possible interactions as DNA gyrase inhibitors. All the synthesized coumarin acetohydrazide compounds have been characterized by spectral analysis IR, 1H NMR and mass spectroscopy. The compounds have been evaluated for In vitro antibacterial and antifungal activity against S. aureus, B. subtilus, E. coli, P. aeruginosa and fungi C. albicans and A. niger. In case of Gram positive bacteria and Gram negative bacteria Compound P5C, M5C, C5C exhibited significant activity. Compounds P5N, M5N, C5N shown moderate activity. Compound P5C, M5C, C5C shown significant Antifungal activity against C. albicans and A. niger. Compounds P5C, M5C, C5C are found to exert significant antibacterial as well as antifungal activity and can serve as potential compound against infectious diseases in future.

Design, synthesis and antibacterial, antifungal activity of some coumarin acetohydrazide derivatives

Aim and Objectives: The aim of the current study is to isolate the starch from elephant foot yam (Amorphophallus paeonifolius) and investigate its potential as a disintegrant in tablet formulation as compare to standard corn starch. The objective of the study is to explore the applications of natural resources and develop an alternative to commercially available starches. Materials and Methods: Starch was isolated by a simple method, evaluated for phytochemical and physico-chemical properties. Tablets were prepared by wet granulation by varying concentrations of elephant foot yam or corn starch in the range of 2.5%, 5%, 7.5% and 10%. Further granules were evaluated for flow properties and tablets were evaluated for post-compression parameters. Results: It was found that the pH of the isolated starch sample was found to be neutral; it exhibited good swelling capacity and fair flow properties. P-XRD pattern showed a C-type diffraction pattern, SEM studies indicated that starch granules had a smooth surface. Granules possessed good flow properties and tablets complied with standard limits of weight variation. Hardness and friability were found in the range of 4.11-4.69 kg/cm2 and 0.11-0.50% respectively. The wetting time was found in the range of 7 to 35 sec for elephant foot yam starch and 16-49 sec for corn starch. Disintegration time for elephant foot yam starch was found to be 28 to 84 sec and for corn starch, it was 40 to 90 sec. Conclusion: Formulations containing elephant foot yam starch showed a similar dissolution profile as that of corn starch. Stability studies were performed on F4 batch and it was found stable for three months.

Exploration of elephant foot yam (Amorphophallus paeoniifolius) starch: An alternative natural disintegrant for pharmaceutical application

Aims: Nanoparticles are the colloidal carrier systems for delivery of poorly soluble drugs. Budesonide. (BUD) a corticosteroid practically insoluble in water is used in asthma treatment. The aim of the present research work was to develop and evaluate BUD nanoparticles. Methodology: The prepared formulation was analyzed for % encapsulation efficiency, particle size analysis, zeta potential, polydispersity index (PDI), scanning electron microscopy and transmission electron microscopy. Poloxamer-188 was found in stabilizing BUD nanoparticles. Results: The observed % encapsulation efficiency of the optimized batch was (82.95) %, particle size was 271.8 nm with PDI 0.456. Solvent injection method was successfully implemented to developed BUD nanoparticles poloxamer-188. Sonication time and amplitude played an important role in governing the particle size. Conclusion: It can be inferred from the study that nanoparticles are a potential drug delivery method for poorly water-soluble drug delivery which can not only get impacted by formulation variables but also by process variables.

Drug administration, flexibility, patient com-pliance, palatability, toxicity are the factors that are required to be considered during the development of pediatric preparations. To overcome the drawbacks of conventional drug delivery systems, the researchers have focused on various innovative methods such as novel drug delivery systems (NDDS). Advancements in Formulation Approaches to Pediatric Oral Drug Delivery Systems

Drug development plays an important role in patient safety and effectiveness. The therapeutic suitability of a new drug depends on the solubility. The solubility of the sparingly soluble drug remains a problem in identifying new active compounds. Solubility plays an important role in achieving optimal drug concentration. Low solubility is not only a concern for the production of formulations, but also an obstacle from the outset when identifying active chemicals for therapeutic purposes. Due to its simplicity in terms of ease of administration and economy, the oral route is the preferred route of drug administration over other routes. Effective aqueous solubility is the first prerequisite for oral medication, since low solubility has poor absorption and bioavailability and unpredictable toxicity of the gastrointestinal mucosa. To avoid these crises, different methodologies are used to improve the solubility and bioavailability of poorly soluble drugs and hydrotropic solubilization is one of them. Hydrotropic agents have the potential to improve the solubility of water-insoluble drugs. In this review, we try to address hydrotropic solubilization methodologies. Hydrotropic Solubilization: An Emerging Approach

The selective optimization of side activities (SOSA) approach appears to be a promising strategy for lead generation. In this approach old drugs are used to generate new hits or leads. The objective of SOSA is to prepare analogues of the hit molecule in order to transform the observed ``side activity{''} into the main effect and to strongly reduce or abolish the initial pharmacological activity. The idea of taking a molecule with a primary activity in humans and then enhancing a secondary effect through structural changes describes the most common implementation of SOSA. An advantage to starting a drug discovery program with molecules that have already been tested in humans is that those molecules have already satisfied many safety criteria. Such molecules also likely have favourable pharmacokinetic profiles. In the present review different successful examples of SOSA switches are summarized. We hope that the present review will be useful for scientists working in the area of drug design and discovery.
