Three dimensional quantitative structure activity relationship (3D QSAR) investigations were carried out on a series of 5-cyano-N1,6-disubstituted 2-thiouracil derivatives for their locomotor activity. The structures of all compounds were built on a workspace of VlifeMDS3.5 molecular modeling software and 3D QSAR models were generated by applying a partial least square (PLS) linear regression analysis coupled with a stepwise variable selection method. Both derived models were found to be statistically significant in terms of regression and internal and external predictive ability (r(2) = 0.9414 and 0.8511, q(2) = 0.8582 and 0.6222, pred_r(2) = 0.5142 and 0.7917). The QSAR models indicated that both electrostatic and steric interaction energies were contributing significantly to locomotor activity of thiouracil derivatives.

3D QSAR investigations on locomotor activity of 5-cyano-N1,6-disubstituted 2-thiouracil derivatives

Despite many herbs having both antidiabetic as well as antiobesity activity; there is no marketed polyherbal formulation for obesity associated diabetes. In light of this, polyherbal preparation for obesity associated diabetes containing Gymnema sylvestre R., Garcinia Cambogia, Lagerstromia speciosa L., was investigated in normal and obese streptozotocin induced diabetic rats. Effect of oral administration of polyherbal formulation (412, 825 and 1625 mg/kg body weight) for 21 days on the level of serum glucose, total cholesterol, triglycerides, serum LDL, serum HDL, serum VLDL levels and body weight in obese diabetic rats were evaluated. Administration of the formulation for 21 days significantly decreased serum glucose, total cholesterol, triglycerides, LDL, VLDL levels and body weight and increased the HDL level. A comparison was made between the action of polyherbal formulation and glibenclamide (4 mg/kg), the standard antidiabetic drug, sibutramine (5mg/kg), the standard antiobesity drug. The antidiabetic and antiobesity effect of the formulation was found to be nearly similar to that observed for glibenclamide and sibutramine respectively. It ican be concluded that, the formulation should be considered as an excellent candidate for future studies of obesity associated diabetes.

Effect of polyherbal formulation in obesity associated diabetes

Cassia tora Linn. is one of the well-known anthraquinone containing plant and has been used in Chinese and Ayurvedic medicine. In the present review, an attempt has been made to explore a literature survey on its traditional uses, phytochemical studies and pharmacological properties. The whole plant as well as specific parts such as roots, leaves and seeds have been widely used and claimed against different diseases by rural and traditional practitioners of Satpura region of Madhya Pradesh. This plant has great contribution in modern system of herbal medicine for new drug development.

Phytochemical and pharmacological profile of Cassia tora Linn. - An Overview

The present study investigated the protective effect of the methanolic extract of fruit of Emblica officinalis (MEO) against indomethacin induced enterocolitis in male Wistar rats. Enterocolitis was induced by subcutaneous administration of indomethacin solution (7.5 mg kg-1, 2 days) for 2 days. The study comprised of 5 groups (n = 6), normal saline-treated, enterocolitis induced, MEO treated groups (100 and 200 mg kg-1, p.o.) and sulfasalazine treated (500 mg kg-1, p.o.) group. On the fourth day animals were sacrificed by cervical dislocation and dissected open to remove gastrointestinal tract. Ileum and colon pieces (10 cm long) are assessed for inflammation on macroscopic score and histopathological studies. Pretreatment with MEO has shown a decreased in macroscopic scores for inflammation as compared to indomethacin treated group. A significant decrease in serum lactate dehydrogenase level was also observed in MEO treated groups. Histopathology examination of MEO treated group revealed less damage compared to indomethacin treated group. The finding of present study provides evidence that Emblica officinalis may be beneficial in patients with inflammatory bowel diseases. {\textcopyright} 2010 Academic Journals Inc.

Effect of Emblica officinalis methanolic fruit extract on indomethacin induced enterocolitis in rats

Simple, accurate, precise and sensitive spectrophotometric method for simultaneous estimation of Montelukast (MON) and Levocetirizine (LEV) in combined tablet dosage form have been developed and validated. The ratio derivative spectroscopic method involves measurement of first derivative amplitude of ratio spectra at 250.4 nm for MON and 238.4 nm for LEV as two wavelengths for estimation. Beer's law is obeyed in the concentration range of 4-12 and 2-6 $\mu$g/mL for MON and LEV, respectively. LOD values for MON and LEV are found to be 0.09 $\mu$g/mL and 0.178 $\mu$g/mL, respectively. LOQ values for MON and LEV are found to be 0.277$\mu$g/mL and 0.591 $\mu$g/mL, respectively. The results of analysis have been validated statistically and recovery studies carried out in the range 80-120% to confirm the accuracy of the proposed method.

Simultaneous determination of montelukast sodium and levocetirizine dihydrochloride in pharmaceutical preparations by ratio derivative spectroscopy

The rate-limiting step to absorption of drugs from the gastrointestinal tract is often dissolution from the dosage form. Allopurinol is a commonly used drug in the treatment of chronic gout or hyperuricaemia associated with leukaemia, radiotherapy, anti-neoplastic agents. One of the major problems with allopurinol is that, it is practically insoluble in water, which results in poor bioavailability after oral administration. In the present study, solid dispersions of allopurinol were prepared by solvent evaporation method, kneading method, co-precipitation method, co-grinding method and closed melting method to increase its water solubility. In the present study amphiphilic carrier like gelucire 50/13 was used in the ratio of 1:1, 1:2 and 1:4. Prepared solid dispersions were characterized in the liquid state by phase solubility studies and in the solid state by Differential Scanning calorimetric analysis, Powder X-ray diffractometry and Fourier Transform Infrared spectroscopy. The aqueous solubility of allopurinol was preferential by the presence polymer with increasing concentration. Solid state characterizations indicated that allopurinol was present as an amorphous material and entrapped in polymer matrix. Mathematical modeling of in vitro dissolution data indicated the best fitting with Korsemeyer-Peppas model and the drug release kinetics primarily as Non-Fickian diffusion. Preparation and in vitro evaluation of allopurinol-gelucire 50/13 solid dispersions

A simple, precise, rapid and accurate RP-HPLC-PDA method has been developed for the simultaneous estimation of drotaverine HCl (DRT) and aceclofenac (ACE) in combined tablet dosage form. Formulation containing DRT with ACE are used as antispasmodic and anti-inflammatory agent. The chromatographic separation was achieved on Qualisil BDS C8 column (250 mm × 4.6 mm, 5.0 $\mu$ particle size) using methanol: THF: acetate buffer (68:12:20 v/v) pH adjusted to 5.0 with acetic acid, flow rate was 1.0 mL min-1 and column was maintained at 50°C. Quantification and linearity was achieved at 298 nm over the concentration range of 1 - 150 $\mu$g mL-1 for Drotaverine HCl and 1.25 - 187.5 $\mu$g mL-1 for Aceclofenac. The method was validated for specificity, linearity, accuracy, precision, LOD, LOQ and robustness. Development and validation of a RP-HPLC-PDA method for simultaneous estimation of drotaverine and aceclofenac in a combined dosage form

Metoprolol succinate (MS) gastroretentive (GR) controlled release system was formulated to increase gastric residence time leading to improved drug bioavailability. Box-Behnken model was followed using novel combinations of sodium alginate (SA), sodium carboxymethylcellulose (NaCMC), magnesium alumino metasilicate (MAS) as independent variables. Floating lag time (Flag), t25, t50, t75, diffusion exponent as dependent variables revealed that the amount of SA, NaCMC and MAS have a significant effect (p < 0.05) on t25, t50, t75 and Flag. MSGR tablets were prepared and evaluated for mass, thickness, hardness, friability, drug content and floating property. Tablets were studied for dissolution for 24 h and exhibited controlled release of MS with floating for 16 h. The release profile of the optimized batch MS01 fitted first-order kinetics (R2 = 0.9868, n = 0.543), indicating non-Fickian diffusion or anomalous transport by diffusion and swelling.

Development and optimization of metoprolol succinate gastroretentive drug delivery system

An ethnobotanical survey was carried out on the use of medicinal plants in Satpuda region of Dhule and Jalgaon districts of Maharashtra. The information was gathered from Pawara, Bhil and Pardhi tribes using an integrated approach of botanical collections, group discussions and interviews with questionnaire during 2004-2005. Among 50 informants interviewed, 8 were tribal practitioners. A total of 67 medicinal plant species distributed in 37 families are documented. In most of the cases, fresh part of the plant was used for the preparation of medicine. These tribal people still depend on the medicinal plants to cure their diseases and disorders in Satpuda forest region. The documented ethnomedicinal plants were mostly used to cure wound infections, skin infections, stomachache, fever, cough, diabetes, diuretics, diarrhoea, eye infections and general weakness.

Use of medicinal plants among tribes in Satpuda region of Dhule and Jalgaon districts of Maharashtra-An ethnobotanical survey

The purpose of this research was to formulate and characterize solid dispersion (SD) of metformin hydrochloride using methocel K100M as the carrier by the solvent evaporation and cogrinding method. The influence of drug polymer ratio on drug release was studied by dissolution tests. Characterization was performed by fourier transform spectroscopy (FTIR), ultraviolet, differential scanning calorimetry and X-ray powder diffractometry. The optimized formulation was subjected to accelerated stability testing as per ICH guidelines. Release data were examined kinetically. SD with 1:4 and 1:5 ratio of drug to polymer obtained by solvent evaporation and cogrinding were selected as the best candidates suitable for prolonged-release oral dosage form of metformin.
