An efficient synthesis of 1-H indazoles

THE FUTURE OF PRESCRIPTION: E-PRESCRIPTION

The fast, economical, precise and highly accurate HPTLC method was developed for the determination of Valdecoxib from its bulk drug and pharmaceutical preparation. Specificity and robustness of the method was studied. Densitogram was used for representing separation of degradation product from Valdecoxib. In this method chromatography was performed on pre-coated silica gel 60 F 254 HPTLC plates (Merck # 5548). Mobile phase used was toluene-ethyl acetate, 5.0+5.0(v/v). Plates were evaluated by densitometry at $\lambda$ = 262 nm with a Camag Scanner II, in conjunction with CATS software for quantitation. The developed method can be used for the quality control and assay of the drug it requires approximately 45min for analysis.

Determination of valdecoxib from its bulk drug and pharmaceutical preparations by HPTLC

This study describes the feasibility of transdermal controlled administration of Losartan potassium (LP) across human cadaver skin. Study also defines the influence of capsaicin, sex and site of application on permeation characteristics and determined an appropriate animal model for human skin permeability. The permeation of LP of various formulations was studied using Keshary-Chein diffusion cell. Optimized controlled formulation (without capsaicin) released 42.17% (±1.85) of LP in 12 hr whereas treatment formulation (with capsaicin 0.028 % w/v) released 48.94% (±1.71) of LP with significant difference on null hypothesis. Influence of sex showed statistically significant difference for permeation of LP through male and female rats, as well as male and female mice across both the abdominal and dorsal sides of the skin (p<0.05). Similarly statistically significant differences were noted for permeation of LP across male and female mice abdomen-dorsal, but not for male rat abdomen-dorsal and female rat abdomen-dorsal. Furthermore, in-vitro permeation of LP across human skin was compared with the permeation across rat and mice skins. Male rat and male mice dorsal skin was found to have closer permeability characteristics to human than other skin membranes, but the Factor of Difference values were < 3 for all membranes which were used suggesting the membranes are good models for human skin permeability. In conclusion simple transdermal adhesive patches formulations incorporating high molecular weight of LP can deliver a dose in-vivo and proposed model skin membranes can be utilized for future pharmacokineic and toxicokinetic studies as well as metabolism studies of LP.

In-vitro percutaneous absorption of losartan potassium in human skin and prediction of human skin permeability

Objective: Husk of Cocos nucifera was screened for diuretic properties. Method: The diuretic properties of aqueous and alcoholic extract of Cocos nucifera husk were evaluated by determination of urine volume, electrolyte concentration and diuretic potency in male albino rats. Different concentrations of aqueous and alcoholic extract (250 mg/kg, 500mg/kg) were orally administered to hydrated rats and their urine output was immediately measured after 5 h of treatment. Frusemide (10mg/kg) was used as reference drug while normal saline (0.9%) solution was used as control. Result: Both the extract exhibited dose dependent diuretic property. The onset of diuretic action was extremely prompt (within 1 h) and lasted throughout the studied period (upto 5 h). Both extract caused marked increase in Na+, K+ and Cl-level. Conclusion: The result of this experiment suggests that Cocos nucifera husk extract possessed significant diuretic activity.

Diuretic activity of Cocos nucifera husk in rats

Effect of alcoholic extract of roots of Rubia cordifolia was studied on elevated blood glucose level in alloxan treated animals. The extract reduced the blood sugar level raised by alloxan. Effect of alcoholic extract was also investigated on cold restraint induced stress and on scopolamine-induced memory impairment. Alcoholic extract enhanced brain $\gamma$-amino-n-butyric acid (GABA) levels and decreased brain dopamine and plasma corticosterone levels. Acidity and ulcers caused due to cold restraint stress were inhibited by alcoholic extract. Animals treated with alcoholic extract spent more time in open arm in elevated plus maze model. It also antagonized scopolamine induced learning and memory impairment. Baclofen induced catatonia was potentiated by alcoholic extract.

Antihyperglycemic, antistress and nootropic activity of roots of Rubia cordifolia Linn

Roots of the plant Inula racemosa are used as folk medicine in east Asia and Europe. Inula racemosa in combination with Commiphora mukul was reported to cure myocardial ischemia. However, systematic investigation of the plant for its specific role in heart diseases has not been conducted so far. In the present study, we have reported the isolation of four major constituents A, B, C and D along with some minor constituents from the plant Inula racemosa. Among the major constituent, constituent Dhas been selected first from spectral data and studied for its cardiac activity on isolated frog heart. The experimental data show that constituent D decreases heart rate and force of contraction at 40 mcg/ml. Actions of Adrenaline are blocked by constituent D and it also acts as an agonist for Propranolol. The studies indicate that constituent D produces a negative ionotropic and negative chronotropic effect on frog's heart. These studies can be utilized as a cardiac marker for exploring the cardiac activity of the plant Inula racemosa. Copyright {\textcopyright} by The Haworth Press, Inc. All rights reserved.

Cardiac activity of isolated constituents of Inula racemosa

Aqueous extract of leaves of M. oleifera was investigated and rationalised for its wound healing activity. The aqueous extract was studied at dose level of 300 mg/kg body weight using resutured incision; excision and dead space wound models in rats. Significant increase in wound closure rate, skin-breaking strength, granuloma breaking strength, hydroxyproline content, granuloma dry weight and decrease in scar area was observed. The prohealing actions seem to be due to increased collagen deposition as well as better alignment and maturation. From the results obtained, it may be concluded that the aqueous extract of M. oleifera has significant wound healing property.

Evaluation of aqueous leaves extract of Moringa oleifera Linn for wound healing in albino rats

A simple, accurate and sensitive spectrophotometric method has been developed for the estimation of rofecoxib in commercial preparations. The lactone ring in the drug is converted to hydroxy acid by sodium carbonate. 1,4-Napthoquinosulphonic acid acts as oxidizing agent and abstracts hydrogen ion from the hydroxy acid to form brown colored chromogen measured at the wavelength of maximum absorption 440 nm against reagent blank. The chromogen obeyed linearity over 20 to 120 $\mu$g/ml (r=0.9998). The method is satisfactorily applied for the analysis of pharmaceutical preparations containing rofecoxib. The results of the analysis were validated statistically and by recovery studies. The recovery ranged between 98.6 and 101.8%.

Spectrophotometric estimation of rofecoxib in tablet dosage forms

Two simple, accurate, rapid and sensitive methods have been developed for the estimation of sildenafil citrate in health care medicines. The method A is based on reduction of ferric ions to ferrous ions by the drug, which further in presence of potassium ferricyanide as oxidizing agent produces blue colored complex measured at 715 nm against reagent blank. The chromogen obeyed linearity over the range of 10 to 70 $\mu$g/ml of drug. The method B is based on reduction of ferric ions to ferrous ions by the drug, which further in presence of potassium dichromate as oxidizing agent produces greenish blue colored complex measured at 700 nm against reagent blank. The chromogen obeyed linearity over the range of 50 to 130 $\mu$g/ml of drug.
