HYPOGLYCEMIC EFFECT OF METHANOLIC EXTRACT OF CITRULLUS LANATUS SEEDS.

In this chapter authors have discussed the role of plants to develop contaminant free environment. This concept is also known as Phytoremediation. Phytoremediation is a word formed from the Greek prefix "phyto" meaning plant and the Latin suffix "remedium" meaning to clean or restore. This technology has been receiving attention lately as an innovative, cost-effective alternative to the more established treatment methods used at hazardous waste sites. Phytoremediation can be classified into different applications, such as phytofiltration or rhizofiltration, phytostabilization, phytovolatilization, phytodegradation and phyto-extraction etc. The chapter will deal with phytoremediation, its advantages, limitations and in detail techniques of classification and application.

Phyto-remediation: Using plants to clean up soils: Phyto-remediation

Background: Ayurvedic medicines and nutraceuticals are gaining popularity among physicians and patients for better therapeutic value. Lack of quality standards and problems, in preparing or testing them, are the main hurdles experienced by both the practitioners and the patients. Objective: The objective of the study was to improve the palatability of the Ayurvedic Nutraceutical Preparation (ADS) by masking its bitter taste and to standardize the taste masking procedure. In the present study Eudragit E 100 was used as an acid soluble coating material. Materials and Methods: ADS powder was converted into granules with PVP K30 as a granulating agent and the ADS granules were coated with Eudragit E 100 coating solution by pan coating process. Various IPQC tests namely flow properties, moisture content were performed on the granules before and after coating for determination of endpoint of granulation and coating respectively. The ADS powder and granules were evaluated for bitter taste. Results: ADS granules were advantageous over ADS powder since the flow properties of ADS granules were better than the flow properties of ADS powder, a prerequisite of pan coating process. Eudragit E 100 inhibited the contact in between the plant extracts and the taste buds due to insolubility of Eudragit E 100 in saliva. Sensory evaluation of taste indicated that the taste of coated granules was significantly masked. Conclusion: The bitter taste of ADS was improved successfully with Eudragit E 100 as a coating agent and the pan coating process. An attempt was made to standardize the pan coating process.

Taste masking of ayurvedic nutraceutical formulation by pan coating process

Impurity profiling has gained importance in modern pharmaceutical analysis due to the fact that an unidentified, potentially toxic impurity are hazardous to health and in order to increase the safety of drug therapy, impurities should be identified and determined by selective methods. Thus a simple sensitive high performance thin layer chromatographic (HPTLC) method for estimation of Strontium ranelate and its impurities has been developed. Strontium ranelate and its impurities were separated and identified on silica Gel 60 F254TLC plates with Acetonitrile: Methanol: Ethyl acetate: Formic acid 9:1:1:1 (v/v) as mobile phase. The quantification was performed at $\lambda$ = 320 nm. The method was validated for linearity, accuracy, precision and robustness according to ICH guidelines. The calibration plots were linear in the range 4000-9000 and 40-90 ng/band for API and its two impurities respectively. The developed method can be used as a quality control tool for Strontium ranelate impurity profile study.

Development and validation of a HPTLC method for determination of strontium ranelate in the presence of its impurities

Biophytum sensitivum (L.) DC (family: Oxalidaceae) has been used in the Indian indigenous system of medicine, Ayurveda, for the treatment of various health aliments including renal calculi. The present study was undertaken to investigate the anti-urolithiatic activity of standardized methanolic extract of whole plant of B. sensitivum (MBS) in rats. Urolithiasis was induced by surgical implantations of zinc disc in the urinary bladders of rats. Upon postsurgical recovery, different doses of MBS (viz., 100, 200 and 400 mg/kg body weight) were administered to zinc disc-implanted rats for the period of 7 days by the oral route. Anti-urolithiatic activity was evaluated by measuring various dimensions of stones and estimating levels of various biomarkers in serum and urine samples. A significant decrease in urinary output was observed in the disc-implanted animals, which was prevented by the MBS treatment. Supplementation with MBS caused significant improvement in glomerular filtration rate and protein excretion. The elevated levels of serum creatinine, uric acid and blood urea nitrogen were also prevented by the MBS treatment. The MBS treatment showed reduced formation of deposition around the implanted zinc disc. The higher dose of MBS (400 mg/kg) found more effective. These results indicate that the administration of MBS significantly prevents the growth of urinary stones. The possible mechanism underlying this effect is mediated collectively through diuretic, antioxidant and anti-inflammatory effects of the plant. The results concluded that the methanolic extract of whole plant of B. sensitivum possessed significant anti-urolithiatic activity.

Anti-urolithiatic activity of standardized extract of Biophytum sensitivum against zinc disc implantation induced urolithiasis in rats

Aim: The present investigation was carried out with an aim to formulate and evaluate praziquantel (PZQ) tablets using solid dispersion approach. Methodology: The solid dispersion was prepared by solvent evaporation method using carriers such as mannitol, urea and PEG 6000 with drug: Carrier ratio of 1:1, 1:2 and 1:3. The solid dispersion was evaluated for physical parameters such as angle of repose, bulk density, Carr′s index, Hausner ratio, drug content and in vitro drug release studies. PZQ tablets (100 mg drug) were prepared further from solid dispersions using direct compression technique. Results: The results of individual assays of solid dispersions with different ratios revealed that the 1:2 ratio of PZQ with PEG 6000 showed higher dissolution rates when compared to others. Tablets compressed were evaluated for their physical parameters such as weight variation, thickness, hardness, friability, dissolution and disintegration tests and compared with plain drug and marketed formulation (Biltricide 600 mg Bayer HealthCare Pharmaceuticals). In vitro dissolution profile of optimized batch (F8) showed better release. The highest solubility was shown by tablet prepared from solid dispersion with 1:2 ratio of PZQ: PEG 6000, which was found to be more than plain drug. Infrared spectra showed that the functional groups of PZQ and PEG 6000 were preserved. Results of 2 3 factorial designs affect the dependent variables such as hardness, disintegration time and % friability. Conclusion: It was concluded that by adopting a systematic formulation approach one can reach to an optimum level. Hence, solid dispersion formulation using PEG 6000 carriers was found to be a good alternative approach for increasing the dissolution rate of PZQ tablets in distilled water.

Design and statistical optimisation of praziquantel tablets by using solid dispersion approach

Background . Oxybenzone, a broad spectrum sunscreen agent widely used in the form of lotion and cream, has been reported to cause skin irritation, dermatitis and systemic absorption. Aim . The objective of the present study was to formulate oxybenzone loaded microsponge gel for enhanced sun protection factor with reduced toxicity. Material and Method . Microsponge for topical delivery of oxybenzone was successfully prepared by quasiemulsion solvent diffusion method. The effects of ethyl cellulose and dichloromethane were optimized by the 3 2 factorial design. The optimized microsponges were dispersed into the hydrogel and further evaluated. Results . The microsponges were spherical with pore size in the range of 0.10–0.22 µ m. The optimized formulation possesses the particle size and entrapment efficiency of 72 ± 0.77 µ m and 96.9 ± 0.52%, respectively. The microsponge gel showed the controlled release and was nonirritant to the rat skin. In creep recovery test it had shown highest recovery indicating elasticity. The controlled release of oxybenzone from microsponge and barrier effect of gel result in prolonged retention of oxybenzone with reduced permeation activity. Conclusion . Evaluation study revealed remarkable and enhanced topical retention of oxybenzone for prolonged period of time. It also showed the enhanced sun protection factor compared to the marketed preparation with reduced irritation and toxicity.

Formulation and Evaluation of Optimized Oxybenzone Microsponge Gel for Topical Delivery

Oxalis corniculata Linn (Oxalidace) is an Indian medicinal plant which is widely used as food and folk medicine. Present work was aimed to check the potential of Oxalis corniculata Linn leaf extract in the treatment of ulcerative colitis. Dried and powdered leaves were extracted using petroleum ether, chloroform, ethyl acetate and methanol. The extracts at a dose of 100 mg/kg were screened for acetic acid induced ulcerative colitis in mice. The antioxidant potential of extracts was evaluated by DPPH method. It was observed that liver and kidney functions were not affected in mice after administration of ethyl acetate and methanol extract for fifteen days. Treatment with ethyl acetate extract of Oxalis corniculata Linn showed superior antiulcer, antioxidant activity and better protective effect compared to other extracts. Myeloperoxidas and malondialdehyde level in blood and tissue was decreased significantly after administration of ethyl acetate and methanol extracts in mice.

Potential of Oxalis corniculata Linn in the treatment of ulcerative colitis

A series of novel indazole derivatives have been designed and synthesized by a novel method and evaluated for their antibacterial activity. The compounds were designed to study their interactions as inhibitor of DNA gyrase B. Salicylaldehyde and hydrazine hydrate have been refluxed along with polyethylene glycol and catalytic amount of ptoluenesulfonic acid to yield desired compounds. All the synthesized compounds were subjected to in vitro anti-bacterial activity studies against B. Subtilis, S. aureus, E. coli, P. Aeruiginosa and S. typhi. Substituted compounds I5, I9 and I13 exhibited significant inhibition of bacterial growth. The MIC value of these 3 compounds was found to be 50$\mu$g/mL. Compounds substituted at 5th and 6th position of indazole were found to show better hydrogen bond interactions with DNA gyrase B and appreciable potential for anti-bacterial activity. They can be used as potential molecule in bacterial resistance cases. Design, Synthesis and Evaluation of Antibacterial Activity of Novel Indazole Derivatives

Increase in complication and expenses involved in marketing of new drug entities has focused greater attention in development of sustained release drug delivery system of existing drugs. NSAID are having short biological half life of 4-4.3 hrs and fluctuation in drug plasma concentration. NSAID are used in symptomatic treatment of rheumatoid arthritis & spondylitis. They have analgesic, anti inflammatory, antipyretics activity. Aceclofenac sustained released tablets was developed using wax and polymer matrix in the different ratio. The evaluation of granules was carried for angle of repose, Hausner's ratio, Carr's compressibility index. Tablets were evaluated for hardness, uniformity of weight, content of active ingredient, thickness, in-vitro dissolution studies. The results of hardness showed that as the concentration of wax or polymer increases, the hardness of tablet increases. The in-vitro dissolution study showed that for the formulation containing Aceclofenac with highest concentration of wax and polymer showed maximum sustained activity. Formulation containing bees wax and Carnauba wax in ratio 1:0.5 were optimized based on in-vitro dissolution and hardness. The dispersion of the drug in the wax and polymer network altered its dissolution profile at pH 6.8, thus making it possible to obtain a gradual and prolonged release and to modulate the release pattern.
