Objective . Objective of the present work was to develop site-specific gastroretentive drug delivery of Troxipide using polymers Pluronic F127 and Polyox 205 WSR. Troxipide is a novel gastroprotective agent with antiulcer, anti-inflammatory and mucus secreting properties with elimination half-life of 7.4 hrs. Troxipide inhibits H. pylori -derived urease. It is mainly absorbed from stomach. Methods . 3 2 factorial design was applied to study the effect of independent variable. Effects of concentration of polymer on dependant variables as swelling index, hardness and % drug release were studied. Pluronic F127 and Polyox 205 WSR were used as rate controlled polymer. Sodium bicarbonate and citric acid were used as effervescent-generating agent. Results . From the factorial batches, it was observed that formulation F5 (19% Pluronic F127 and 80% Polyox 205 WSR) showed optimum controlled drug release (98.60% ± 1.82) for 10 hrs with ability to float &gt;12 hrs. Optimized formulation characterized by FTIR and DSC studies confirmed no chemical interactions between drug and polymer. Gastroretention for 6 hrs for optimized formulations was confirmed by in vivo X-ray placebo study. Conclusion . Results demonstrated feasibility of Troxipide in the development of gastroretentive site-specific drug delivery.

Design and Evaluation of Polyox and Pluronic Controlled Gastroretentive Delivery of Troxipide

Objective: The present research work was undertaken with an aim to formulate a combination dosage form constituting an antacid and analgesic which may be useful in case of hyperacidity associated headache. Methods: In this formulation, a mixture of magnesium and aluminium hydroxide was used as the antacid. Ibuprofen was used as analgesic drug while, citric acid and tartaric acid along with sodium bicarbonate were used as effervescent mixture. The prepared tablets of all the formulations were evaluated for physical characterization. Results: The infra red spectra revealed that there was no chemical interaction between the drug and excipients which showed their compatibility. The results of these studies were found to be within the standard Pharmacopoeial limits. The release character was studied using the disintegration and dissolution studies conducted. The results of 23factorial design revealed that the amounts of crosspovidone, tartaric acid and citric acid used in effervescent formulation significantly affected the dependent variables i. e. Hardness, friability and disintegration time etc. The stability studies of the tablet formulation showed that the tablets remained stable even after exposing to stress condition of temperatures. Conclusion: It was thus concluded that by adopting a systematic formulation approach, optimized release mechanism can be reached in the shortest time with minimum efforts.

A new concept in nanotechnology is that of Solid lipid nanoparticles (SLNs), i.e. lipid nanocrystals in water possessing a solid core into which drugs are incorporated. The SLNs combine the virtues of more traditional drug carriers such as liposomes or polymeric nanoparticles while eliminating some of their disadvantages, e.g. the issues of burst release and long term stability in the case of liposomes as well as the problems of residual solvents and bulk production in the case of polymeric nanoparticles. The use of nanoparticles as drug-carrier system is a very attractive possibility to achieve controlled drug release. A clear advantage of SLNs over polymeric nanoparticles is the fact that the lipid matrix is made from physiological lipids, which decreases the danger of acute and chronic toxicity. There are various types of solid lipid nanoparticles and can be prepared using several methods. The present review covers all the aspects of solid lipid nanoparticles including their history, types, methods of preparations, evaluation parameters, advantages over conventional drug delivery systems and pharmaceutical applications which may be helpful in carrying out extensive research in their regards.

Solid lipid nanoparticles: A critical appraisal

Objective: Quality is considered to be the major tool in pharmaceutical industry. Failures in the quality of upcoming new chemical entities have been seen in the recent years due to complications in the formulation process. Capecitabine (CTB), a anticancer pro-drug which orally shows fluctuation in plasma drug concentration and serious side effects. Purpose of this study is to understand the effect of variables on quality of the formulation. The aim of the present study was to develop CTB-loaded nano polymeric micelles (PMs) using Quality-by-Design (QbD) approach to screen the effect of eight formulation and process factors on the formulation. Methods: CTB-loaded nano PMs were prepared by organic solvent/water (o/w) emulsion technique. Plackett-Burman (PB) design was used to screen the effect of eight formulation and process factors to improve anticancer efficacy. The nano PMs were characterized by drug content (% DC), entrapment efficiency (% EE), particle size, zeta potential, TEM, PXRD, SEM and in-vitro drug release. Results: Particle Size and zeta potential of nano PMs were found to be in the range of 178 - 1572 nm and -10.9 to -36.1 mV respectively. Analysis of the Pareto chart revealed that the HP $\beta$-CD and Eudragit S100 significantly influences drug content and entrapment efficiency respectively. Nano PMs demonstrated sustained release of CTB. Conclusion: The study concludes that the statistical PB design could be useful to identify influencing variables such as HP $\beta$-CD and Eudragit S100 that can be used for further investigation.

Screening of factors using plackett burman design in the preparation of capecitabine-loaded nano polymeric micelles

A novel bilayered mucoadhesive buccal patch of zolmitriptan was prepared using xanthan gum (XG) as mucoadhesive polymer. Hydroxypropyl methylcellulose E-15 was used as film-former and polyvinyl alcohol (PVA) was incorporated, to increase the tensile strength of the patches. To study the effect of independent variables viz. concentrations of XG and PVA, on various dependent variables like in vitro drug release, ex vivo mucoadhesive strength and swelling index, 32 factorial design was employed. In vitro drug release studies of optimized formulation showed initially, rapid drug release; 43.15% within 15 min, followed by sustained release profile over 5 h. Incorporation of 4% dimethyl sulfoxide enhanced drug permeability by 3.29 folds, transported 29.10% of drug after 5 h and showed no buccal mucosal damage after histopathological studies. In conclusion, XG can be used as a potential drug release modifier and mucoadhesive polymer for successful formulation of zolmitriptan buccal patches. {\textcopyright} 2013 Elsevier B.V. Formulation and in vitro evaluation of xanthan gum-based bilayered mucoadhesive buccal patches of zolmitriptan

Cancer nanotechnology is the latest trend in cancer therapy. It helps the pharmacist to formulate the product with maximum therapeutic value and minimum or negligible range side effects. Cancer is the disease in which the abnormal cells are quite similar to the normal cell with just minute functional or genetic change. Thus, it is very hard to target the abnormal cells by the conventional method of the drug delivery system. Nanotechnology is probably the only method that can be used for site-specific action without causing the side effects by killing the normal cells. This review article describes the possible way to exploit the nanotechnology to targeted drug therapy in cancer. The various methods used are: systemic delivery systems, passive targeting, active targeting, intracellular delivery, subcellular localization, and nanoparticle drugs. Different cancer detection techniques like carbon nanotubes, nanorods, and biosensors are also available. This review article gives an idea about the possible potential of nanotechnology in drug delivery, drug targeting, and the diagnosis of cancer.

A novel green approach by photocatalytic oxidative polymerization was developed that addresses and resolves current limitations of chemical polymerization of anilines. This process generates nanoparticles of the conducting polymer poly(N-ethylaniline) (PNETA) and poly-(N-methylaniline) (PNMTA) in a single step by using a catalytic amount of the oxidant in the presence of an organic acid such as DL-(±)-Tartaric acid. Poly(N-ethylaniline) nanoparticles doped with tartaric acid show exceptional antimicrobial activity at low concentration. Five compounds have been synthesized. Of these, two compounds have been found to exhibit significant activity against Klebsiella pneumoniae. The minimum inhibitory concentration value was observed at 50 g/mL. An extremely simple green approach is described that generates bulk quantities of nanoparticles with average diameter 200 nm size of the conducting polymer PNETA in one-step photochemical method using a catalytic amount of ammonium persulfate as oxidant in the presence of organic acid dopant, DL-(±)-Tartaric acid. The resultant polymer were characterized by UV-visible, FTIR, XRD and SEM analysis and their antibacterial activity against Klebsiella pneumoniae has been studied. These polymer nanomaterials show a prominent antimicrobial activity against the tested bacteria and therefore have potential applications in biomedical sciences. {\textcopyright} 2014 Taylor and Francis.

Study of biological activity of conducting poly(n-ethylaniline) nanoparticles doped with organic acid

OBJECTIVE\nTo investigate neuroprotective and antioxidant effect of Phoenix dactylifera (P. dactylifera) (PD) fruits. \n\nMETHODS\nMethanolic extract of P. dactylifera fruits (MEPD) at doses of 30, 100 and 300 mg/kg was studied against permanent BCCAO (long-term hypoperfusion) in rats. Chronic occlusion of bilateral common carotid arteries (BCCA) caused significant elevation in malondialdehyde levels due to increased lipid peroxidation as well as decrease in levels of other biochemical enzymes i.e. glutathione, glutathione peroxidase, glutathione reductase, glutathione-S-transferse, catalase and superoxide dismutase. \n\nRESULTS\nPost occlusion treatment for 15 d with 100 and 300 mg/kg doses of MEPD significantly reduced the enhanced malondialdehyde levels and reversed the alterations in the declined levels of antioxidant enzymes in brain homogenates of hypoperfused rats. Long-term cerebral hypoperfusion in rats caused a propensity towards anxiety and restlessness (open field paradigm) accompanied by deficits of spatial learning and memory (Morris water maze testing). Additionally, histopathological observations in hypoperfused brains revealed reactive changes like shrinkage and necrosis of neurons. 100 and 300 mg/kg doses of MEPD significantly alleviated these alterations. \n\nCONCLUSIONS\nThese results confirmed the protective role of P. dactylifera in ischemia hypoperfusion and thereby it's beneficial role in cerebrovascular insufficiency states and related complications.

Neuroprotective and antioxidant role of Phoenix dactylifera in permanent bilateral common carotid occlusion in rats

Tablet coating is the key step involved in the manufacturing of tablets having controlled release, delayed release profiles. The tablet coating have number of advantages like masking odor, taste, color of the drug, providing physical and chemical protection to drug, Protecting drug from the gastric environment. Tablets are usually coated in horizontal rotating pan with coating solution is either directly poured or sprayed on to them. The amount of coating on the surface of a tablet is critical to the effectiveness of the oral dosage form. Recent trends in tablet coating focuses on overcoming disadvantage of solvent based coating. This review concerns with the coating process, equipments involved, coated tablets evaluation and specialized coating techniques. Tablets are usually coated in horizontal rotating pans with the coating sprayed onto the free surface of the tablet bed. Tablets must have a coating mass that lies within a prescribed range with very little inter-and intra-tablet coating variability. Using the Discrete Element Method (DEM) tablet coating can be simulated on the computer. Simulation data provide the position, velocity and orientation of each tablet within the coater allowing accurate measurements of the time and orientation that each tablet spends exposed to the coating spray.
