This study was undertaken to investigate the possible effect of hydroalcoholic root extract of Rubia cordifolia against indomethacin-induced enterocolitis in rats. Male Wistar rats received vehicle or hydroalcoholic root extract of Rubia cordifolia (300 and 600 mg/kg) for 11 consecutive days. Enterocolitis was induced by subcutaneous administration of indomethacin (7.5 mg/kg) on 8 th and 9 th day. The colonic mucosal injury was assessed by macroscopic scoring and histopathological examination. Furthermore, the serum lactate dehydrogenase activity was estimated. Indomethacin treatment to rats produced acute intestinal inflammation, manifested by a thickening of the bowel wall, mesenteric haemorrhage, mesentery adhesion and multiple mucosal ulcers of small intestine and colon. Treatment with hydroalcoholic root extract of Rubia cordifolia revealed less damage to intestinal tissue and decreased serum lactate dehydrogenase activity which was elevated by induction of colitis. The present data suggests protective effect of Rubia cordifolia in indomethacin-induced enterocolitis and may be beneficial in patients with inflammatory bowel diseases.

Evaluation of anti-leukemic activity of potentised Catharanthus roseus : An in-vitro study

In the present work the effect of two different diluents namely dicalcium Phosphate (DCP) and maize starch (MS) on the release of aceclofenac from sustained release matrix tablets has been studied. Gum damar (GD), a hydrophobic polymer with significant binding property was used as a release retarding agent. In the present study aceclofenac was used as a model drug for developing sustained release matrix tablets owing to its short biological half life (4 hours) and dosing frequency more than one tablet per day. Matrix tablet of aceclofenac using synthetically prepared hydrophilic polymers (e.g. HPMC) have been reported. However, low cost, ease of availability and non toxic property makes use of natural gums (e.g. gum damar), an alternative approach for development of sustained release matrix tablets. Compatibility between aceclofenac and GD was studied using FTIR. Sustained release matrix tablets of aceclofenac using GD as release retardant were prepared by wet granulation technique. Matrix tablets were prepared using different strengths of GD (10% - 30% w/w) with respect to total tablet weight and two different excipients DCP & MS as diluents. The tablet weight (400mg) and diameter (10mm) was kept constant. The physicochemical properties such as hardness, thickness, friability, uniformity of weight and the drug content of the formulated tablets were estimated The in vitro dissolution profile of the formulated tablets was compared with marketed sustained release aceclofenac tablets. The kinetics of release of aceclofenac from the matrix tablets were found to follow Hixon Crowell cube root law indicating a surface dependent release. The study revealed that GD as release retardant along with MS as diluent show results comparable to that of the marketed sustained release tablets of aceclofenac.

Effect of two different diluents on release profile of aceclofenac from sustained release matrix tablets using gum damar as release retardant

A simple, economical, precise and accurate method for simultaneous determination of Diclofenac potassium (DICLO K) and Thiocholchiside (THIO) in combined tablet dosage form has been developed. The first method is based on Absorption corrected and second method is Area Under Curve (AUC) Spectrophotometry. The first method was based on the absorption corrected method in which DICLO K and THIO exhibit λmax at 264.99 nm and 373.84 nm, respectively in methanol. Quantitative estimation of THIO was carried out by subtracting the absorption due to DICLO K at 333.74 nm using experimentally calculated absorption factor and wavelength ranges of 252.56-260.59 nm and 278.51-285.53 nm were selected to determine DICLO K and THIO by AUC method respectively in combined formulations. Beer's law is obeyed in the concentration range of 25-75 μg mL-1 for DICLO K and 4-12 μg mL-1 for THIO for both the methods. The % assay for commercial formulation was found to be in the range 99.01 - 100.89 % for DICLO K and 98.91 - 101.72 % for THIO by the proposed methods. These methods were validated with respect to linearity, precision and accuracy as per ICH analytical method development guidelines. Recovery was found in the range of 98.16 - 100.02 % for DICLO K and 98.56 - 99.81 % for THIO by absorption corrected method and 98.56-99.87% for DICLO K and 99.68-100.21% for THIO by AUC method respectively.

Spectrophotometric simultaneous determination of diclofenac potassium B.P. and thiocolchicoside in combined tablet dosage form by absorption corrected method and area under curve method

Sodium bisulfite has been reported first time for the synthesis of 2,5-disubstituted 1,3,4-oxadiazole using microwave and conventional method in ethanol-water. The yields obtained are in the range of 90-95% using microwave and 87-91% using conventional method. All the synthesized compounds (8a-8s) are novel and were evaluated for their in vitro antifungal activity. SAR for the series has been developed by comparing their MIC values with miconazole and fluconazole. Some of the compounds from the series like 8k was equipotent with miconazole against Candida albicans and Fusarium oxysporum. Microwave assisted one pot synthesis of some novel 2,5-disubstituted 1,3,4-oxadiazoles as antifungal agents

Diltiazem hydrochloride is a calcium channel blocker generally indicated for the treatment of angina and hypertension and it is extensively metabolized due to the hepatic metabolism. Formulation of diltiazem hydrochloride into an oro-dispersible dosage form can provide fast relief with higher bioavailability. The bitter taste of the drug should be masked to formulate it in a palatable form. In the present work, an attempt was made to mask the taste by complexation technique, with a formulation into an oro-dispersible dosage form, using superdisintegrants Doshion P544, crospovidone (CP) and sodium starch glycolate (SSG). The complexes of diltiazem hydrochloride with $\beta$-CD (1:1 molar ratio) were prepared by kneading, co-evaporation, co-grounding, freeze-drying and melting methods. Phase solubility showed stability constant 819.13M-1. Prepared inclusion complexes were evaluated for taste masking and characterized by I.R, XRD, DSC. Using the drug $\beta$-CD complex, oro-dispersible tablets were prepared and evaluated for hardness, friability, weight variation, thickness, disintegrating time (DT), dissolution rate and taste. Formulations with 4% Doshion, 8% CP and 4% SSG showed DT of 0.54, 0.35 and 1.23 minutes, respectively.

Formulation and in vitro evaluation of taste-masked oro-dispersible dosage form of diltiazem hydrochloride

Aceclofenac, nonsteroidal anti-inflammatory drug, the bioavailability of aceclofenac is very low which is mainly due to its low solubility in water. Phase solubility diagram of aceclofenac in aqueous $\beta$-cyclodextrin solution were constructed with concentration 0-15 mMole. The phase solubility diagram of aceclofenac with $\beta$-cyclodextrin could be classified as A L-type, indicating 1:1 stochiometry. Binary inclusion complex of aceclofenac with $\beta$-cyclodextrin was prepared by kneading and spray drying technique, physical mixture was prepared for comparison. Effect of water soluble polymer (poly ethylene glycol 6000) was studied on kneaded binary kneaded inclusion complex in two proportions 5 % and 10 %. Binary and ternary systems of aceclofenac with $\beta$-cyclodextrin prepared by various methods were characterized by differential scanning calorimetry and Powder X-ray diffractometry. It could be concluded that aceclofenac could form inclusion complex with $\beta$-cyclodextrin. The dissolution rate of aceclofenac was increased by inclusion complexation with $\beta$-cyclodextrin. The ternary kneaded complex with 10% poly ethylene glycol shows improvement in dissolution rate as compared to kneaded inclusion complex. The ternary kneaded complex (10% poly ethylene glycol) and spray dried inclusion complex exhibits fast dissolution as compared to plain aceclofenac. In phramcodynamic study, spray dried complex and ternary kneaded complex (10% poly ethylene glycol) significantly decreases acetic acid induced writhing as compared to control group and thus exhibited significant analgesic activity, whereas aceclofenac was found to be insignificant. {\textcopyright} RJPT All right reserved.

Preparation and characterization of ternary inclusion complex of aceclofenac-β-cyclodextrin

Tinidazole is a broad spectrum antiprotozoal agent having bitter taste. The present study concerns to mask the bitter taste of tinidazole as an orally disintegrating tablet (ODT) using Indion 294 and Indion 204 (ion exchange resin) as a taste masking agent. In first method, 1:1, 1:2 and1:3 ratios of drug and resin were stirred for six hour. Percent drug losses were found to be on higher side in this method. In second method, drug-resin complexes were prepared by wet granulation method with different concentration ratios of drug with Indion 294 as well as with Indion 204(1:0.5, 1:1,1:1.5) using lactose as diluent and polyvinylpyrrolidone k-30 as binder. On the basis of disintegration time, optimum concentration of superdisintegrants i.e. crosscarmollose sodium and crosspovidone were found to be 8% w/w. The tablets were prepared by direct compression method using orange flavour and evaluated for taste, weight variation, hardness, friability, disintegration test, content uniformity, dissolution study &amp; FTIR study. The best formulations were found to be containing (1:1.5) drug: Indion 204 &amp; (1:1.5) drug: Indion 294.

Formulation and evaluation of taste masked orally disintegrating tablet of tinidazole

The process of understanding the control and capability (PUCC) is an iterative closed loop process for continuous improvement. It covers the DMAIC toolkit in its three phases. PUCC is an iterative approach that rotates between the three pillars of the process of understanding, process control and process capability, with each iteration resulting in a more capable and robust process. It is rightly said that being at the top is a marathon and not a sprint. The objective of the six sigma study of Ranitidine hydrochloride tablets is to achieve perfection in tablet manufacturing by reviewing the present robust manufacturing process, to find out ways to improve and modify the process, which will yield tablets that are defect-free and will give more customer satisfaction. The application of six sigma led to an improved process capability, due to the improved sigma level of the process from 1.5 to 4, a higher yield, due to reduced variation and reduction of thick tablets, reduction in packing line stoppages, reduction in re-work by 50%, a more standardized process, with smooth flow and change in coating suspension reconstitution level (8%w/w), a huge cost reduction of approximately Rs.90 to 95 lakhs per annum, an improved overall efficiency by 30% approximately and improved overall quality of the product.
