A reliable, selective and sensitive stability-indicating RP-HPLC assay was established for the quantitation of bromazepam (BMZ) and one of the degradant and stated potential impurities; 2-(2-Amino-5-bromobenzoyl) pyridine (ABP). The assay was accomplished on a C18 column (250 mm × 4.6 mm i.d., 5 ?m particle size) and utilizing methanol-water (70: 30, v/v) as the mobile phase, at a flow rate of 1.0 ml min-1. HPLC detection of elute was obtained by a photodiode array detector (DAD) which was set at 230 nm. ICH guidelines were adhered for validation of proposed method regarding specificity, sensitivity, precision, linearity, accuracy, system suitability and robustness. Calibration curves of BMZ and ABP were created in the range of 1 16 ?g mL-1 with mean recovery percentage of 100.02 1.245 and 99.74 1.124 and detection limit of 0.20 ?g mL-1 and 0.24 ?g mL-1 respectively. BMZ stability was inspected under various ICH forced degradation conditions and it was found to be easily degraded in acidic and alkaline conditions. The results revealed the suitability of the described methodology for the quantitation of the impurity (ABP) in a BMZ pure sample. The determination of BMZ in pharmaceutical dosage forms was conducted with the described method and showed mean percentage recovery of 99.39 1.401 and 98.72 1.795 (n = 6), respectively. When comparing the described procedure to a reference HPLC method statistically, no significant differences between the two methods in regard to both accuracy and precision were found.

Development and validation of a stability indicating RP-HPLC-DAD method for the determination of bromazepam

Mashi is a black colored powder formulation obtained after combustion of the plant or animal drug. It is prepared by bahirdhum padhati (outside) or anterdhum padhati (in the close vessel). In this dosage form, bulk of raw material is reduced to a greater extent by the application of a certain quantum of energy. Due to this treatment, hidden chemical constituents become prominent and/or a new chemical moiety is formed which is therapeutically active. This formulation is cost-effective and easy to prepare. This review article aims to highlight the different mashi formulations mentioned in Ayurvedic text and also incorporate the formulation not mentioned in the Ayurvedic text but used by Ayurvedic practitioners. The objective was to introduce researchers to the simple yet excellent formulation mashi which should be studied in detail to establish its identity, purity, and therapeutic activity. © 2021 The Authors

A review through therapeutic attributes of Ayurvedic formulation mashi

Unfavorable side effects of available antipsychotics limit the use of conventional delivery systems, where limited exposure of the drugs to the systemic circulation could reduce the associated risks. The potential of intranasal delivery is gaining interest to treat brain disorders by delivering the drugs directly to the brain circumventing the tight junctions of the blood–brain barrier with limited systemic exposure of the entrapped therapeutic. Therefore, the present research was aimed to fabricate, optimize and investigate the therapeutic efficacy of amisulpride (AMS)-loaded intranasal in situ nanoemulgel (AMS-NG) in the treatment of schizophrenia. In this context, AMS nanoemulsion (AMS-NE) was prepared by employing aqueous-titration method and optimized using Box-Behnken statistical design. The optimized nanoemulsion was subjected to evaluation of globule size, transmittance, zeta potential, and mucoadhesive strength, which were found to be 92.15 nm, 99.57%, −18.22 mV, and 8.90 g, respectively. The AMS-NE was converted to AMS-NG using poloxamer 407 and gellan gum. Following pharmacokinetic evaluation in Wistar rats, the brain Cmax for intranasal AMS-NG was found to be 1.48-folds and 3.39-folds higher when compared to intranasal AMS-NE and intravenous AMS-NE, respectively. Moreover, behavioral investigations of developed formulations were devoid of any extrapyramidal side effects in the experimental model. Finally, outcomes of the in vivo hematological study confirmed that intranasal administration of formulation for 28 days did not alter leukocytes and agranulocytes count. In conclusion, the promising results of the developed and optimized intranasal AMS-NG could provide a novel platform for the effective and safe delivery of AMS in schizophrenic patients. © 2021 Elsevier B.V.

Nose-to-brain delivery of amisulpride-loaded lipid-based poloxamer-gellan gum nanoemulgel: in vitro and in vivo pharmacological studies

Background: Over two billion people around the world suffer from anemia. Majority of populations are using dietary supplements and herbal medicines for the management of the anemic conditions. Many polyherbal formulations such as Raktavardhak Kadha (RK), are available in the Indian market as a nutritional supplement and herbal-based medicine for anemia. Objectives: The present study is aimed at investigating antianemic potential of polyherbal formulation, RK, against phenylhydrazine-induced anemia in rats. Materials and methods: RK was subjected to preliminary phytochemical analysis and iron estimation. Anemia was induced by phenylhydrazine administration (40 mg/kg, i.p.) for 2 consecutive days. Antianemic activity of RK was investigated at the dose of 1.8 ml/kg, twice daily for 12 days by estimating blood parameters and pathological changes in liver, heart, spleen and bone marrow. Results: RK was found to contain saponins, steroids, flavonoids, tannins and phenolic compounds. Iron content was found to be 5 mg/100 ml in RK. Anemia induction by phenylhydrazine injections to rats caused significant decrease in red blood cells (RBCs), hemoglobin and hematocrit. These decreased levels of RBCs, hemoglobin and hematocrit in blood was significantly improved by the treatment with RK. Furthermore, RK restored pathological changes in liver, heart, spleen and bone marrow tissues near to normal. Conclusion: This study suggests antianemic activity of RK, which can be attributed to its iron content and ability to prevent hemolysis.

Post injury, healing of wound is essential for recovery of uprightness of the body, which is one of the complex, continuous and unanticipated chains of events in case of diabetic patients. Nitric oxide represents a potential wound therapeutic agent due to its ability to regulate inflammation and eradicate bacterial infections. Impaired wound healing is a prominent diabetic complication which may lead to amputations also. In addition to modern medicines we can use nitric oxide therapy prominently for diabetic wound healing. Prominent and proven role of nitric oxide as well as conventional materials (like metformin and hydrogen sulphide, whey proteins, acidified nitrile etc), therapies (like low level laser therapy, hyperbaric oxygen therapy etc) and techniques (like in vivo implants with biosensors) can be taken into consideration. Many plant extracts showed promising results for wound healing activity by increasing nitric oxide levels. Use of modern technologies such as implant with biosensor and technique like sonic head hog gene are available for diabetic wound healing using Nitric oxide. In this review, an attempt has been made to compile comprehensive updated information of role of nitric oxide in diabetic wound healing, which may be exploited by focusing more on development of effective strategies to treat diabetes-associated wound.

Breast cancer is the form of cancer most prevalent and intensified progressively among the women population. The propagation of breast cancers takes place in different stages and diagnosed lately. Various approaches have been emerged to treat this clinical condition but these are also integrated with varied side effects. The reason might be attributed to undesired effects of the chemotherapeutic agent and/or haphazard damage to both healthy and cancerous cells. These hitches induce the urge for targeting cancerous cells by the utilization of novel therapeutic platforms. Nano-drug delivery systems are a cluster of different approaches to treating various severe diseases. Henceforward this concept is also applied in the treatment of breast cancer. Nanoparticles exhibits numerous benefits mainly, reduction in dose and low toxicity, solubility enhancement of certain drugs, increased cellular uptake etc. These are the efficient carrier of hydrophobic drugs as these drugs possess challenges in solubility and bioavailability. Cellular uptake by tumor cells is better by nanocarriers owing to a smaller size. Nanocarriers For Breast Cancer: Advanced Perspective [Meme Kanseri İçin Nanotaşıyıcılar: Gelişmiş Perspektif]

General Principles, Types, Diagnosis and Management of Poisoning

Spatial arrangement for cells and the opportunity thereof have implications in cell regeneration and cell proliferation. 3D inverse space (3DIS) substrates with micron-sized pores are fabricated under controlled environmental conditions from polymers such as poly(lactic-co-glycolic) acid (PLGA), poly(lactic acid) (PLA) and poly(styrene) (PS). The characterization of 3DIS substrates by optical microscopy, scanning probe microscopy (SPM), etc. shows pores within 1-18 $\mu$m diameter and prominent surface roughness extending up to 3.9 nm in height over its base. Conversely, to compare two-dimensional (2D) versus 3DIS substrates, the crucial variables of cell height, cell spreading area and cell volume are compared using lung adenocarcinoma (A549) cells. The results indicate an average cell thickness of ∼6 $\mu$m on a glass substrate whereas cells on PLGA 3DIS were ∼12 $\mu$m in height, occasionally reaching 20 $\mu$m, with a 40% decreased cell spreading area. A549 cells cultured on polymer 3DIS substrates show a cell regeneration growth pattern, dependent on the available spatial volume. Furthermore, PLGA 3DIS cell culture systems with and without graded doxorubicin (DOX) pre-treatment result in potent cell inhibition and cell proliferation, respectively. Additionally, standard DOX administration to A549 cells in the PLGA 3DIS system revealed altered drug sensitivity. 3DIS demonstrates utility in facilitating cellular regeneration and mimicking cell proliferation in defined spatial arrangements.
