Ramipril is an ACE inhibitor mainly used for management of mild to severe hypertension and myocardial infarction. The poor solubility and wettability of Ramipril leads to poor dissolution and hence showing variations in bioavailability. The present study is aimed to increase solubility and dissolution of the drug using solid dispersion techniques. The formulations were characterized by solubility studies, X-Ray Diffractometry studies, differential scanning calorimetry, Fourier transform infrared spectroscopy and in vitro dissolution rate studies. The solubility of drug increased linearly with increase in polymer concentration showing AL type solubility diagrams. The solid dispersions of the drug demonstrated higher drug dissolution rates than physical mixtures and pure Ramipril. The kneaded complex method showed higher dissolution rate than other complex method and it was incorporated into buccal Patches. The patches were prepared by solvent casting method using HPMC K15 and Poloxomer WSR205NF. The patches were found to be smooth in appearance, uniform in thickness, weight uniformity, drug content, swelling index, folding endurance, surface pH and in vitro diffusion study using Keshery chien diffusion cell. The Patch of 0.5% HPMC K15 exhibit in vitro release of 78.16% through cellophane membrane and 72.16% release through egg membrane and the patch of 0.5% Poloxomer WSR205 NF exhibit in vitro release of 74.24% through cellophane membrane and 69.44% release through egg membrane in 8 hrs showing good mucoadhesive strength and mucoadhesive time. The optimised patch was subjected to ex vivo studies through goat buccal mucosa showed 60.47% release in 8 hrs.

Improvement of dissolution rate of Ramipril by solid dispersion technique and devlopement of buccal patch

Simple, precise, rapid and accurate methods for simultaneous determination of Lamivudine, Zidovudine and Nevirapine in combined tablet dosage form have been developed. Method is based on Ratio spectra derivative and methanol is used as solvent. The amplitudes at 229.77 nm, 316.40 nm and 341.76 nm of the first derivative of ratio spectra were selected to determine Lamivudine, Zidovudine and Nevirapine, respectively by ratio derivative method. Beer's law is obeyed in the concentration range of 6-18 $\mu$g/ml, 12-36 $\mu$g/ml and 8-24 $\mu$g/ml for Lamivudine, Zidovudine and Nevirapine, respectively by Ratio Derivative method. The % assay in commercial formulation was found to be in the range 99.01 - 100.90 % for Lamivudine, 99.3 - 101.11% for Zidovudine and 98.91 - 101.72 % for Nevirapine by the proposed method. The method was validated with respect to linearity, precision and accuracy. Recovery was found in the range of 99.36 - 100.23 % for Lamivudine, 98.91 - 101.32% for Zidovudine and 99.0 - 100.4 % of Nevirapine by ratio derivative method. The method developed is simple, economical, precise and accurate and can be used for routine quality control of combined tablets.

Simultaneous estimation of Lamivudine, Zidovudine and Nevirapine by ratio derivative spectroscopic method

Two simple, economical, precise and accurate methods are described for the simultaneous determination of Dexketoprofen (DKP) and Thiocolchicoside (TCS) in combined tablet dosage form. The first method (Method A) is Absorption Corrected Method and second method (Method B) is First Order Derivative Spectrophotometry. The amplitudes at 300.0 nm and 373.45 nm in the Absorption Corrected Method and 244.14nm and 268.022nm in the first order derivative Spectrophotometry were selected to determine DKP and TCS, respectively in combined formulation. The methods were validated by following the analytical performance parameters suggested by the International Conference on Harmonization (ICH). All validation parameters were within the acceptable range. Under experimental conditions described, calibration curve, assay of tablets and recovery studies were performed. A critical evaluation of proposed methods were performed by statistical analysis of data where slope, intercept, correlation coefficient is shown in Table (1). As per the ICH guidelines, the method validation parameters checked. Beer's law is Obeyed in the concentration range of 6.25-75 TimesNewRoman,Italic.-1.c01g/ml for DKP and 1-12 TimesNewRoman,Italic.-1.c01g/ml for TCS by both the methods.

Spectrophotometric Simultaneous Determination of Dexketoprofen and Thiocholchicoside in Combined Tablet Dosage Form by Absorbance Corrected Method and First Order Derivative Method

Three simple, economical, precise and accurate methods are described for the simultaneous determination of Mefenamic acid (MFA) and Paracetamol (PCT) in combined tablet dosage form. The first method (Method A) is Dual wavelength, second method (Method B) is Area under curve and third method (Method C) is Absorption Corrected In the First method, MFA was determined by plotting the difference in absorbance at 234.73 and 260.70 nm (difference is zero for PCT) against the concentration of MFA. Similarly for the determination of PCT, the difference in absorbance at 243.5 and 267.79 nm (difference is zero for MFA) was plotted against the concentration of MFA in combined formulation. In second method wavelength ranges of 233.017-236.34 nm and 267.79-271.0 nm were selected to determine PCT and MFA by AUC method in combined formulation. In the third method MFA concentration was determined directly from calibration plot by measuring absorbance at 343 nm and PCT was determined after correction for absorbance of MFA at 233 nm. The methods were validated by following the analytical performance parameters suggested by the International Conference on Harmonization (ICH). Beer's law is obeyed in the concentration range of 5-25 $\mu$g/mL for MFA and 4.5-22.5 $\mu$g/mL for PCT by both the methods.

Spectrophotometric simultaneous determination of mefenamic acid and paracetamol in combined tablet dosage form by dual wavelength, area under curve and absorbance corrected method

The aim of the current study was the development of Esomeprazole buccal adhesive tablets using direct compression. Buccoadhesive tablets of Esomeprazole which were designed to prolong the buccal residence time after oral administration. Buccal adhesive formulations were developed using with various combinations of mucoadhesive polymers namely, Carbopol 934 P, HPMC K4M, sodium alginate. The prepared Esomeprazole tablets were evaluated for weight variation, thickness, hardness, surface PH, content uniformity and swelling index. In-vitro drug Permeation Studies by egg membrane, ex-vivo drug Permeation Studies by goat membrane. The effect of formulation composition on the release pattern was also investigated. Most formulations showed Esomeprazole controlled release profiles depended on the grade and polymer ratio. Tablets containing Carbopol 934 P and sodium alginate in the ratio of 1:1(B3) showed the maximum drug release of 94.16% in 8 hours. The swelling index was proportional to sodium alginate content and inversely proportional to Carbopol 934 P content. Formulation B3 showed maximum swelling index after 6 hours. The surface pH of all tablets was found close to neutral PH. The release mechanisms were found to fit Peppas' kinetic model. {\textcopyright} RJPT All right reserved.

Controlled release from directly compressible esomeprazole buccal tablet

The aim of the present study was to study effect of Xanthan gum and Chitosan in combination on effervescent floating matrix tablet of water soluble analgesic drug. Tapentadol hydrochloride. Tapentadol hydrochloride is a synthetic opioid used as a centrally acting analgesic and effective in both experimental and clinical pain. The half-life of the drug is about 4 hours and oral dose is 50 to 250 mg twice a day. To reduce the frequency of administration and to improve patient compliance, a sustained-release formulation of Tapentadol is desirable. The 32 full factorial design was employed to study effect of Xanthan gum and Chitosan in combination on Tapentadol hydrochloride floating tablets. Sodium bicarbonate was incorporated as a gas-generating agent. Combination of polymers Xanthan gum and Chitosan was used to retard drug release. The concentration of polymers was varied and their effect on floating time, drug content, % drug release after 8 hours, swelling index and hardness of the tablets was studied. The formulation was evaluated using Infrared-red spectroscopy and Differential Scanning Calorimetry to study drug-excipient compatibility. From the factorial batches, it was observed that formulation containing combination of 20% sodium bicarbonate and 10% citric acid shows optimum floating ability whereas the formulation containing 20% Xanthan gum and 28% Chitosan shows optimum sustained drug release pattern. X ray study of the optimized formulations showed gastro retention for 6 hrs indicating successful floating GRDDS of Tapentadol HCl.

Design, development and evaluation of floating tablets of tapentadol hydrochloride using Chitosan

Clonazepam (CLZ) is an anticonvulsant benzodiazepine widely used in the treatment of epilepsy. CLZ is a BCS Class II drug and its bioavailability is thus dissolution limited. The objective of the present study was to prepare solid dispersions (SDs) of CLZ by various techniques, using the amphiphilic carrier Gelucire 50/13 in various proportions, to increase its water solubility. Drug-polymer interactions were investigated by Fourier-transform infrared (FTIR) and Ultra-Violet (UV) spectroscopy. The SDs were characterized physically by differential scanning calorimetry (DSC) and X-ray diffraction (XRD). A phase solubility study was performed and the stability constant (Ks) was found to be 275.27, while the negative Gibbs free energy ($\Delta$G otr) indicated spontaneous solubilization of the drug. The dissolution study showed that the SDs considerably enhanced the dissolution rate of the drug. The FTIR and UV spectra revealed no chemical incompatibility between the drug and Gelucire 50/13. XRD patterns and the DSC profiles indicated the CLZ was in the amorphous form, which explains the improved dissolution rate of the drug from its SDs. Finally, mouth dissolving tablets (MDTs) were prepared from the optimized batches (kneading method) of solid dispersion, using crospovidone and Doshion P544 resin as superdisintegrants. The tablets were characterized by in-vitro disintegration and dissolution tests. The study of the MDTs showed disintegration times in the range 32.0±0.85 to 20.0±1.30 sec and dissolution was faster than for the commercial preparation. In conclusion, this investigation demonstrated the potential of solid dispersions of a drug with Gelucire 50/13 for promoting the dissolution of the drug and contributed to the understanding of the effect of a superdisintegrant on mouth dissolving tablets containing a solid dispersion of a hydrophobic drug.

Solubility enhancement and formulation of mouth dissolving tablet of Clonazepam with solid dispersion technology

Three simple, economical, precise and accurate UV Spectrophotometric methods have been developed for the estimation of Mycophenolate sodium in bulk and pharmaceutical formulations. Method A involves measurement of absorbance of solution at 250.23 nm in methanol. Method B involves measurement of integrated area under curve in the wavelength range of 248.07 nm - 250.03 nm and is used to construct calibration curve. Method C is first order derivative spectroscopy method, in which first order derivative amplitude at 258.42 nm is measured. Linearity for detector response was observed in the concentration range of 10 - 50 $\mu$g/ml for all the three methods in methanol. The results of analysis have been validated as per ICH analytical method validation guidelines. Accuracy, precision and recovery studies were carried which confirmed the validity of the proposed methods. {\textcopyright} RJPT All right reserved.

Development and Validation of UV Spectrophotometric Methods for Estimation of Mycophenolate Sodium in Bulk and Pharmaceutical Formulations

A simple, precise, rapid and accurate high-performance thin-layer chromatographic (HPTLC) method has been developed for the simultaneous estimation of thiocolchicoside (THIO) and aceclofenac (ACE) in bulk and pharmaceutical dosage form on silica gel precoated aluminum 60F 254 plates (10 cm × 10 cm) with 250 mm thickness. The separation was carried out in methanol-chloroform-water 9.6:0.2:0.2 (v/v/v) as mobile phase. The densitometric scanning was carried out at 254 nm. The R F values were found to be 0.70 ± 0.05 for THIO and 0.83 ± 0.05 for ACE. The linearity was obtained in the range 30-180 and 750-4500 ng per band for THIO and ACE with correlation coefficients r 2 = 0.9995 and 0.9993, respectively. The method was validated for specificity, linearity, accuracy, precision, limit of detection (LOD), limit of quantification (LOQ) and robustness. The percentage recovery for both the analytes was in the range of 98.7-101.2%. Development and validation of an HPTLC method for simultaneous estimation of thiocolchicoside and aceclofenac in combined dosage form

Aim: The present study was undertaken to investigate antiurolithiatic property of aqueous extract of fruit of Terminalia chebula in Wistar albino rats. Materials and methods: The protective effect of aqueous extract of Terminalia chebula fruit was evaluated at two dose levels (100 and 200 mg/kg body weight) using ethylene glycol induced calcium oxalate urolithiasis model in rats. Results: The results indicate that ethylene glycol treatment decreases calcium level in urine and increased that in the kidney tissue homogenate, which were prevented in animal receiving simultaneous treatment of extract. Extract treatment decreased the elevated levels of oxalate and phosphate in urine as well as kidney tissue homogenate. The extract supplementation also prevented the elevation of serum levels creatinine, uric acid and blood urea nitrogen. Histopathological study revealed that extract reduced histological changes and retained the normal architecture of kidney tissue. Conclusion: This finding indicates that aqueous extract of Terminalia chebula fruit possessed antiurolithiatic activity. {\textcopyright} 2012 Elsevier Masson SAS. All rights reserved.
