The ophthalmic in-situ gels now days proved an palpable sustained drug delivery in various eye diseases. The formulation of in-situ gels for eye which carries the advantages like easy for administration, reduces frequency of dose and improves patient compliance. The formation of in-situ gels depends on phase transition system or sol-gel transition system. The formulation approaches like temperature intonation, pH change and presence of ions from which the drug gets released in a sustained and controlled manner are utilised for in-situ gels. Various polymers that are used for the formulation of in-situ gels include chitosan, Pluronic F-127, poly-caprolactone, gellan gum, alginic acid, xyloglucan, pectin etc. INTRODUCTION: Ophthalmic drug delivery is one of the challenging endeavors facing the pharmaceutical scientist today. The structural and functional aspects of the eye render this organ highly impervious to foreign substances. A significant challenge to the formulator is to overcome the protective barriers of the eye without causing permanent tissue damage. The major problem encountered with topical administration is the rapid pre-corneal loss caused by nasolacrymal drainage and high tear fluid turnover which leads to only 10% drug concentrations available at the site of actions 1 .

A Novel Ophthalmic Drug Delivery System: in-Situ Gel

Simple, sensitive, selective, precise stability indicating chromatographic methods for determination of Drotaverine Hydrochloride and Diclofenac Potassium from pharmaceutical tablets were developed and validated as per the ICH guidelines. A novel stabilityindicating LC assay method was developed and validated for quantitative determination of Drotaverine and Diclofenac in bulk drugs and in pharmaceutical dosage form. An isocratic, reversed phase LC method was developed using an Qualisil BDS C8 column (250 mm × 4.6 mm, 5.0 $\mu$ particle size), using methanol: THF: acetate buffer (45:08:17 v/v) pH adjusted to 5.0 with acetic acid mobile phase, flow rate was 0.7 mL min -1 and column was maintained at 50 °C. The detection was carried out at 292 nm and injection volume was 20 $\mu$L. The peak purity was checked with the photodiode array detector. Drotaverine Hydrochloride and Diclofenac Potassium were subjected to acid, alkali and neutral hydrolysis, oxidation, photo degradation and dry heat and wet heat treatment. Degradation products were well separated from the analytes peaks. As the method could effectively separate the drug from its degradation products, it can be employed as stability-indicating one. The developed method was validated with respect to linearity, accuracy (recovery), precision, specificity and robustness.

Development and validation of a stability-indicating lc method for simultaneous analysis of drotaverine hydrochloride and diclofenac potassium and its application for formulation analysis

A simple, rapid and precise reversed-phase liquid chromatographic method is developed for simultaneous determination of Aliskiren and Hydrochlorothiazide in their binary mixture of commercial pharmaceutical preparations. This method, reported first time for a binary mixture, uses a Kromasil C18, 250 × 4.6 mm, 5 $\mu$m analytical column. Analytes were estimated by isocratic elution with acetonitrile (ACN): tetrahydrofuran (THF):water (71: 13: 16% v/v) mobile phase (pH adjusted to 3 with ortho phosperic acid) at flow rate of 0.5 mL/min; the column maintained at 50°C and detector was set at 229 nm. The sample concentrations are measured on weight basis to avoid the internal standard. The method is validated and shown to be linear. The correlation coefficients for aliskiren and hydrochlorothiazide are 0.9998 and 0.9994, respectively. The recovery values for aliskiren and hydrochlorothiazide ranged from 99.25-99.54% and 98.84-100.01%, respectively. The relative standard deviation for six replicates is always less than 2%. The method is successfully applied for simultaneous quantitative analysis of the title drugs in combined tablets.

Simultaneous estimation of Aliskiran and hydrochlorothiazide in pharmaceutical formulation by RP-LC-PDA

Simple, precise, rapid and accurate methods for simultaneous determination of Atazanavir (ATV) and Ritonavir (RTV) in combined tablet dosage form have been developed. Method A is based on Ratio spectra derivative and method B uses integrated area under curve (AUC), methanol is used as solvent for both the methods. The amplitudes at 280.01nm and 286.12nm of the first derivative of ratio spectra were selected to determine ATV and RTV, respectively by ratio derivative method and wavelength ranges of 246.97-252.03nm and 240.78-244.16nm were selected to determine ATV and RTV by AUC method in combined formulation. Beer's law is obeyed in the concentration range of 15-75$\mu$g/mL and 5-25$\mu$g/mL for atazanavir and ritonavir, respectively by both the methods. The % assay in commercial formulation was found to be in the range 98.81-100.8% for ATV and 98.91-101.2% for RTV by the proposed methods. The methods were validated with respect to linearity, precision and accuracy. Recovery was found in the range of 99.20-99.96% for ATV and 99.12-99.97% for RTV by ratio derivative method and 99.08-99.66% for ATV and 99.78-100.6% for RTV by AUC method. The methods developed are simple, economical, precise and accurate and can be used for routine quality control of analytes in combined tablets.

Spectrophotometric simultaneous determination of atazanavir and ritonavir in combined tablet dosage form by ratio derivative and area under curve method

Simple, precise and economical spectrophotometric method have been developed for the simultaneous estimation of Metoprolol Succinate (METO) and Amlodipine(AMLO) in combined tablet dosage form. Present study describes the development and validation for the simultaneous estimation of metoprolol(METO) and Amlodipine(AMLO) by the Ratio derivative UV spectroscopy method. Methanol is used as solvent. The amplitudes at 277.017nm and 235.62nm of the first derivative of ratio spectra were selected to determine METO and AMLO, respectively by ratio derivative method. Beer's law is obeyed in the concentration range of 50-250 $\mu$g/mL and 5-25 $\mu$g/mL for Metoprolol Succinate and Amlodipine, respectively by both the methods. The % assay in commercial formulation was found to be 99.56% for METO and 98.96% for AMLO by the proposed method. The method was validated with respect to linearity, precision and accuracy. Recovery was found in the range of 99.51-99.92% for METO and 99.69-100.6% for AMLO by ratio derivative method. The present result shows that the proposed method can be successfully employed for routine analysis of the marketed formulations. {\textcopyright} RJPT All right reserved.

Ratio derivative spectrophotometry method for simultaneous estimation of Metoprolol and Amlodipine in their combined dosage form

A simple, economical, precise and accurate methods for simultaneous determination of Dutasteride (DUTA) and Tamsulosin (TAM) in combined tablet and capsule dosage form has been developed. The first method is based on Ratio Spectra Derivative (Method A) and second method is based on Dual Wavelength Spectrophotometry (Method B). Methanol was used as solvent for both the methods. The amplitudes 250 nm and 234.20 nm in the second derivative of the ratio spectra were selected to determine DUTA and TAM, respectively. In the second method i.e. Dual method in which two wavelengths were selected for each drug in a way so that the difference in absorbance is zero for another drug. At wavelengths 221.43 nm and 227.47 nm tamsulosin have equal absorbance therefore these two wavelengths were used to determine Dutasteride (DUTA) and on similar basis 272.35 nm and 276.05 nm were selected to determine Tamsulosin (TAM) in combined formulation. The drugs obey Beer's law in the concentration range of 5-25 TimesNewRoman, Italic.-1.c01g mL-1 for DUTA and 4-20 TimesNewRoman, Italic.-1.c01g mL-1 for TAM by both the methods. The methods were validated with respect to linearity, precision and accuracy. The results of analysis have been validated statistically and recovery studies confirmed the accuracy and reproducibility of the proposed methods which were carried out by following ICH guidelines. The proposed methods were applied successfully to determine uniformity of contents in commercial capsule and tablet formulations.

Spectrophotometric Simultaneous Determination of Dutasteride and Tamsulosin in Combined Tablet Dosage Form by Ratio Derivative and Dual wavelength Method and its Application to determine Uniformity of Contents

One simple, economical, precise and accurate method is described for the simultaneous determination of Dapoxetine and Sildenafil in combined tablet dosage form. The method is Absorption Corrected Method. The amplitudes at 237.54 nm and 325.92 nm in the Absorption Corrected Method were selected to determine DPT and SL, respectively in combined formulation. The method was validated by following the analytical performance parameters suggested by the International Conference on Harmonization (ICH). All validation parameters were within the acceptable range. Under experimental conditions described, calibration curve, assay of tablets and recovery studies were performed. A critical evaluation of proposed method was performed by statistical analysis of data where slope, intercept, correlation coefficient is shown in Table (1). As per the ICH guidelines, the method validation parameters checked. Beer's law is obeyed in the concentration range of 5-25 $\mu$g/ml for DPT and 8-40 $\mu$g/ml for SL by the given method.

Spectrophotometric simultaneous determination of dapoxetine and sildenafil in combined tablet dosage form by absorbance corrected method

A fast, simple, precise and accurate high performance thin layer chromatographic method was developed and validated for the simultaneous estimation of paracetamol, ibuprofen and caffeine in combined tablet dosage form. The method employed pre-coated silica gel 60F254 plate as stationary phase and a mixture of ethyl acetate: glacial acetic acid (9.5: 0.5 v/v) as mobile phase. The plate was scanned and quantified at 205 nm. The two drugs were satisfactorily resolved with Rf 0.48 for paracetamol, 0.75 for ibuprofen and 0.17 for caffeine respectively. Linearity was found to be in the range of 300-1100 ng/band for paracetamol, ibuprofen and caffeine respectively. Intra-day and inter-day precision in %RSD were found to be 1.21-1.62% for paracetamol, 1.16-1.50% for ibuprofen and 1.29-1.64% for caffeine respectively. The developed method was validated for linearity, accuracy, precision, limit of detection and limit of quantitation, specificity, robustness parameters as per ICH guidelines. The correlation coefficient of paracetamol, ibuprofen and caffeine were found to be 0.9995, 0.9991 and 0.9992 respectively. The average percentage recovery of paracetamol, ibuprofen and caffeine was found to be 100.25%, 99.67% and 99.63% respectively. The proposed HPTLC method has potential applications for routine simultaneous analysis of paracetamol, ibuprofen and caffeine in combined marketed tablet dosage form. © RJPT All right reserved.

Development and validation of HPTLC method for simultaneous estimation of paracetamol, ibuprofen and caffeine in bulk and pharmaceutical dosage form

Three simple, economical, precise and Accurate UV Spectrophotometric methods have been developed for the estimation of Montelukast sodium in bulk and pharmaceutical formulations. Method A involves measurement of absorbance of solution at 344.30 nm in methanol. Method B involves integrating area under curve in the wavelength range of 342.93 nm-343.75 nm and is used to construct calibration curve. Method C is first order derivative spectroscopy method, in which first derivative amplitude at 347.91 nm is measured. Linearity for detector response was observed in the concentration range of 5-25 μg/ml for all the three methods in methanol. The results of analysis have been validated as per ICH analytical method validation guidelines. Accuracy, precision and recovery studies were carried and confirmed the validity of the proposed methods.
