CHEMICAL STANDARDIZATION OF VIDANGA CHURNA FORMULATION AND QUANTITATIVE ESTIMATION OF EMBELIN BY UV SPECTROPHOTOMETRIC METHOD

The immunomodulatory activity of an Indian medicinal plant i.e. ethanolic extract from Dodonaea viscosa L.F. namely DV was studied for there phagocytic activity, cell mediated and humoral immune system on rat/mouse. Immunomodulatory effect was assessed in carbon clearance test, delayed type of hypersensitivity (DTH), T-cell population test and sheep erythrocyte agglutination test (SEAT) in animal treated with DV at doses of 200 and 400 mg/kg. In carbon clearance test, D.viscosa exhibited significantly high phagocytic index against control group, indicating stimulation of the reticulo-endothelial system. Significant decrease in mean difference, in the foot paw thickness in DTH indicates its anti-inflammatory activity. In SEAT D. viscosa treated groups at 200, 400 mg/kg doses in normal immune status animals while In T-cell population test, showed significant increase in T-cell rosette formation against control. These results confirm the immunomodulatory activity of D. viscosa extract, which is a known immunomodulator in indigenous medicine.

Immunomodulatory activity of ethanolic extract of Dodonaea viscosa L.F

This article describes a new, simple, precise and accurate TLC method for simultaneous quantitation of atorvastatin (ATO), ezetimibe (EZE) and fenofibrate (FEN) as the bulk drug and in tablet dosage forms. Chromatographic separation of the drugs was performed on aluminum plates precoated with silica gel 60 F254 as the stationary phase and the solvent system consisting of chloroform-toluene-methanol-acetic acid 6:3:0.5:0.15 ($\upsilon$/$\upsilon$/$\upsilon$/$\upsilon$). Densitometric evaluation of the separated zones was performed at 263 nm. The drugs were satisfactorily resolved with RF values of 0.16 ± 0.02, 0.22 ± 0.02 and 0.76 ± 0.02 for ATO, EZE and FEN, respectively. The accuracy and reliability of the method was assessed by evaluation of linearity (75-375 ng per spot for ATO, 99-594 ng per spot for EZE and 66-330 ng per spot for FEN), precision intra-day and inter-day RSD values were always less than 1.51 for the titled drugs, accuracy (99.72 ± 0.75% for ATO, 101.25 ± 0.91% for EZE and 101.06 ± 0.60% for FEN) and specificity, in accordance with ICH guidelines.

Validated TLC method for simultaneous quantitation of atorvastatin, ezetimibe and fenofibrate in bulk drug and formulations

In present study, antifungal paper soap strips of fluconazole were prepared &amp; evaluated for dermal infections because the presence of thick foam on the infected part causes hydration of stratum corneum for better penetration of drug. The formulation and evaluation of medicated soap strips were carried out in two phases. Phase-I studies optimization, which was carried out to study water absorption capacity using different types of papers (Whatman filter paper no. 1, 41 and 42, filter paper, bond paper and butter paper) by determining the parameters weight gain, size, shape and foam test. From these parameters best soap, concentration of liquid soap solution and papers were selected. Phase-II studies involve incorporation of drug in best soap solution and papers chosen from phase-I studies. The medicated soap strips were prepared by dipping the selected paper in drug containing soap solution. The prepared medicated paper soap strips were characterized for weight gain, foam test, drug content uniformity, FTIR, in vitro drug release studies using diffusion cell with hydrated cellophane membrane and primary skin irritation test. The microbial studies indicate drug activity by zone of inhibition against microorganisms C.albicans, S.typhi, S.aureus, E.coli, klibsiella, B.subtilis, P.aeroginosa, A.niger and A.fumigatus species. The obtained results for prepared medicated soap strip of fluconazole indicate that these strips are economic, convenient, gives good foam on application, uniform in drug content and not showing any skin irritation.

Formulation and evaluation of fluconazole soap strips for dermal infections

A simple, selective, rapid, and precise RP-HPLC-PDA method has been developed for the simultaneous estimation of Lornoxicam (LOR) and Thiocolchicoside (THIO) in pharmaceutical dosage form by reverse phase liquid chroma-tography using Waters Symmetry C18 (250 mm × 4.6 mm, 5.0 μ) column. The mobile phase consisting of metha-nol: THF: acetate buffer (60: 10: 30 v/v); pH adjusted to 5.5 with glacial acetic acid at a flow rate of 0.75 mL min-1 and column was maintained at 50 °C with detection at 382 nm. The retention time of Thiocolchicoside and Lornox-icam was 3.36 and 4.08 minutes, respectively. The method was validated according to the ICH guidelines with re-spect to specificity, linearity, accuracy, precision and robustness, limit of detection and limit of quantification. Li-nearity of Lornoxicam and Thiocolchicoside were in the range of 0.2 to 80 μg mL-1 and 0.1 to 40 μg mL-1, respec-tively and its percentage recovery were found to be 100.37 % and 100.51 %, respectively. The proposed method is suitable for simultaneous determination of Lornoxicam and Thiocolchicoside in pharmaceutical dosage form. Development and validation of a RP-HPLC-PDA method for simultaneous determination of lornoxicam and thiocolchicoside in pharmaceutical dosage form and its application for dissolution study

Bauhinia racemosa Lam. (Caesalpiniaceae) leaves have been used in the treatment of asthma traditionally and we therefore undertook this study to scientifically validate its benefit in asthma using suitable animal models. Antihistaminic principles are known to be useful in the treatment of asthma; hence, in the present work, the antihistaminic activity of an ethanol extract of B. racemosa (at a dose of 50 mg/kg, i.p.) was assessed using clonidine-induced catalepsy and haloperidol-induced catalepsy in Swiss albino mice. The results showed that the ethanol extract inhibits clonidine-induced catalepsy but there is no effect on haloperidol-induced catalepsy. This suggests that the inhibition is through an antihistaminic action and that there is no role of dopamine. Hence, we concluded that the ethanol extract has significant antihistaminic activity. The polar constituents in the ethanol extract of leaves of B. racemosa may be responsible for the antihistaminic activity and B. racemosa may therefore have a role in the treatment of asthma.

Antihistaminic effect of Bauhinia racemosa leaves

Cardiovascular diseases and their treatment pose a great challenge. Many instances of cardiovascular disease occur in the early morning hours. Hence, the objective of this study was to develop a time-controlled release formulation of metoprolol succinate based on a pulsatile multiparticulate (pellets) drug delivery system. The formulation was intended to be administered in the evening at 22:00 hours to evaluate symptoms of cardiovascular disease that are experienced in the early morning hours (from 04:00 to 06:00). Drug layering followed by a swelling layer and finally by an insoluble coat application was done using a Sanmour fluid bed processor. Metoprolol succinate layered on sugar pellets (74% w/w) layered with 20% (w/w) ion doshion resin P-547 and coated with 15% (w/w) ethocel with the addition of 20% castor oil showed a lag time of 4 h and was then followed a sigmoidal release pattern with more than 95% drug having been released by the 10th h.

Pulsatile multiparticulate drug delivery system for metoprolol succinate

Formulation and in vitro evaluation of losartan potassium (LP) loaded transdermal delivery system (TDS) was investigated for controlled release and improved therapeutic efficacy. TDS (patches) were prepared by varying the composition of Eudragit RL 100 and Eudragit RS 100 (5:0, 4:1, 3:2, 2.5:2.5, 2:3, 1:4 and 0:5). Patches were evaluated for thickness, content uniformity, mechanical properties, moisture uptake and in vitro drug release. Technological parameters for all the formulations were found to be within the limit. In vitro studies showed relatively high permeation of LP (F1-42.17 ± 1.13%) from the formulation comprising 4:1 ratio of polymer. Inclusion of capsaicin (55.70 ± 1.55%) and pluronic F-68 (70.88 ± 1.20%) to formulation F1 resulted increased permeation of LP across human skin. In conclusion, this study demonstrated the potential of simple transdermal adhesive patch incorporating LP to deliver therapeutically useful dose in-vivo for the treatment of hypertension.

Studies on in-vitro transcutaneous delivery of losartan potassium, influence of penetration enhancers and barrier membrane

Allopurinol is a commonly used drug in the treatment of chronic gout or hyperuricaemia associated with treatment of diuretic conditions. One of the major problems with the drug is that it is practically insoluble in water, which results in poor bioavailability after oral administration. In the present study, solid dispersions of allopurinol were prepared by solvent evaporation, kneading method, co-precipitation method, co-grinding method and closed melting methods to increase its water solubility. Hydrophilic carriers such as polyvinylpyrrolidone, polyethylene glycol 6000 were used in the ratio of 1:1, 1:2 and 1:4 (drug to carrier ratio). The aqueous solubility of allopurinol was favored by the presence of both polymers. These new formulations were characterized in the liquid state by phase solubility studies and in the solid state by differential scanning calorimetry, powder X-ray diffraction, UV and Fourier Transform Infrared spectroscopy. Solid state characterizations indicated that allopurinol was present as an amorphous material and entrapped in polymer matrix. In contrast to the very slow dissolution rate of pure allopurinol, the dispersion of the drug in the polymers considerably enhanced the dissolution rate. Solid dispersion prepared with polyvinylpyrrolidone showed highest improvement in wettability and dissolution rate of allopurinol. Mathematical modeling of in vitro dissolution data indicated the best fitting with Korsemeyer-Peppas model and the drug release kinetics primarily as Non-Fickian diffusion. Therefore, the present study showed that polyvinylpyrrolidone and polyethylene glycol 6000 have a significant solubilizing effect on allopurinol.

Physicochemical characterization and solubility enhancement studies of allopurinol solid dispersions

The recent research on modified release drug delivery systems gives special attention to pulsatile drug delivery systems because of its unique release pattern. A pulse has to be designed in such a way that a complete and rapid drug release is achieved after the lag time so as to match body's circadian rhythms with the release of drug which increases the efficacy and safety of drugs by proportioning their peak plasma concentrations during the 24 hours in synchrony with biological rhythm. The aim of this review is to describe several types of drug delivery systems. The data include rational behind the use of chronotherpeutic release of drug and application in various diseases and prerequisites of drug for pulsatile drug delivery system. Advantages, disadvantages and commercial marketed technologies of pulsatile drug delivery system launched by pharmaceutical companies is also included in data.
