Ramipril is an ACE inhibitor used for management of hypertension and myocardial infarction. It exhibits poor bioavailability of 28% which is due to its poor solubility and high first pass metabolism. The present work was aimed at overcoming these two limitations. Drug-$\beta$-cyclodextrin and Hydroxy propyl $\beta$-cyclodextrin complexes were prepared and characterized by Fourier Transformation Infrared spectroscopy, Differential Scanning Calorimetry and powder X-Ray Diffractometry studies. In vitro studies showed that the solubility and dissolution rate of Ramipril were significantly improved by complexation with $\beta$-cyclodextrin and Hydroxy propyl $\beta$-cyclodextrin with respect to drug alone. The kneaded complex showed higher dissolution rate than other complex and it was incorporated into buccal Patches. The patches were prepared by solvent casting method using hydroxyl propyl methyl cellulose (HPMC K15) and Poloxomer WSR205NF. The patches were found to be smooth in appearance, uniform in thickness, weight uniformity, drug content, swelling index, folding endurance, surface pH and in vitro diffusion study using Keshery chien diffusion cell. The Patch of 0.5% HPMC K15 exhibit in vitro release of 81.16% through cellophane membrane and 75.72% release through egg membrane and the Patch of 0.5% Poloxomer WSR205 NF exhibit in vitro release of 77.24% through cellophane membrane and 71.08% release through egg membrane in 8 hrs showing good mucoadhesive strength and mucoadhesive time. The Optimized patch was subjected to ex vivo studies through goat buccal mucosa showed 63.49% release in 8 hrs.

Solubility enhancement and formulation of buccal patches of Ramipril cyclodextrin complex

Darifenacin is a urinary antispasmodic. The oral absorption of darifenacin is poor due to its low solubility and poor bioavailability (15–19%). Darifenacin was complexed with hydroxylropyl beta-cyclodextrin (Hp $\beta$ -CD). The best results were obtained with the coevaporation that interacts in a 1 : 1 drug : cyclodextrin molar ratio. The solid inclusion complexes were found to be amorphous in the characterization. The dissolution rate of darifenacin from the Hp $\beta$ -CD solid inclusion complex was increased compared to the powdered drug. The controlled release buccoadhesive patches for the delivery of darifenacin were prepared using HPMC K100M CR and HPMC K15. The coevaporation complex of the drug was used in the formulation due to its increased saturation solubility and increased ease of dissolution. The patches were evaluated for their surface pH, folding endurance, swelling, mucoadhesive properties, in vitro residence time, vapour transmission test and in vitro and ex vivo release studies. Formulations Hb2 (2%) and Pb4 (4%) were found to be optimized. These two formulations can be used for buccal delivery of darifenacin which avoids first pass effect and leads to increased bioavailability of darifenacin.

Dissolution Rate Enhancement, Design and Development of Buccal Drug Delivery of Darifenacin Hydroxypropyl β-Cyclodextrin Inclusion Complexes

A simple and sensitive UV spectrophotometric method was developed and validated for the determination of pregabalin in bulk & pharmaceutical formulations. The method was linear in the range of 6-14 $\mu$g/ml with a correlation coefficient of 0. 9986. There is no generally accepted method for the determination of pregabalin. The absorbance was measured at 210 nm. The method was validated with respect to accuracy, precision, Stability, limit of detection and limit of quantitation. This method is found to be simple, specific, precise, accurate, reproducible and low cost UV Spectrophotometric method.

Development and validation of UV spectrophotometric method of pregabalin in bulk and pharmaceutical formulation

Phoenix dactylifera (PD) has been claimed for its neuroprotective potential in the traditional system of medicine but has not yet been scientifically documented. Phytochemical reports of Phoenix dactylifera fruits have demonstrated the presence of polyphenols and flavonoids, which have already been documented to play major role in neuroprotection against various experimental models of cerebral ischemia/reperfusion. In the present study, we have investigated the neuroprotective as well as antioxidant properties of methanolic extract of Phoenix dactylifera fruits (MEPD) at 30, 100, 300 mg/kg p.o against global cerebral ischemia-induced oxidative stress and neuronal death. The global cerebral ischemia was induced by occlusion of bilateral common carotid arteries for 5 min followed by 24 h of reperfusion. Varied biochemical/enzymatic alterations, produced subsequent to the application bilateral common carotid artery occlusion (BCCAO) followed by reperfusion viz. increase in lipid peroxidation and decrease in glutathione, glutathione reductase, catalase, glutathione-S-transferase, glutathione peroxidase and superoxide dismutase, were markedly reversed and restored to near normal levels in the groups pre-treated with 15 days. The pretreatment also reversed the histopathological changes induced by global cerebral ischemia in CA1 hippocampal region. The protective action, exhibited by MEPD against global cerebral induced brain injury, suggests its therapeutic potential in cerebrovascular diseases (CVD) including stroke. These findings are important because the present treatment strategies for CVD are far from adequate and PD with wide usage is known to be a safe natural product. {\textcopyright} 2013 Elsevier Masson SAS.

Protective effects of Phoenix dactylifera against oxidative stress and neuronal damage induced by global cerebral ischemia in rats

A simple stability-indicating isocratic RP-HPLC method was developed and validated for the determination of mycophenolate sodium and its alkali degradation product. Forced degradation of the drug was carried out under thermolytic, photolytic, acid/base hydrolytic and oxidative stress conditions. Alkali degradation product DP1 was isolated and separation of stress degradation products was achieved on a Symmetry C18 (250 × 4.6 mm × 5.0 $\mu$m) column using the mobile phase methanol-acetate buffer adjusted with acetic acid to pH 6.0 (76 + 24, v/v) at a 0.55 mL/min flow rate and 50°C. Data were integrated at the detection wavelength of 251 nm. The method validation characteristics included accuracy, precision, linearity, range, specificity and sensitivity per International Conference on Harmonization guidelines. Robustness testing was conducted to evaluate the effect of minor changes in the chromatographic conditions and to establish appropriate system suitability parameters. Structural elucidation of degraded products was performed by HPLC/MS/MS. The method was used successfully for drug product analysis, dissolution study and determination of the drug's acid, alkali and oxidative degradation kinetics.

Development and validation of a stability-indicating HPLC-photodiode array detector method for formulation analysis and degradation kinetics and dissolution studies of mycophenolate sodium and HPLC/MS/MS characterization of its stress degradation products

The purpose of the present work was to design and optimize compression coated floating pulsatile drug delivery systems of bisoprolol. Floating pulsatile concept was applied to increase the gastric residence of the dosage form having lag phase followed by a burst release. The prepared system consisted of two parts: a core tablet containing the active ingredient and an erodible outer shell with gas generating agent. The rapid release core tablet (RRCT) was prepared by using superdisintegrants with active ingredient. Press coating of optimized RRCT was done by polymer. A 32 full factorial design was used for optimization. The amount of Polyox WSR205 and Polyox WSR N12K was selected as independent variables. Lag period, drug release and swelling index were selected as dependent variables. Floating pulsatile release formulation (FPRT) F13 at level 0 (55 mg) for Polyox WSR205 and level +1 (65 mg) for Polyox WSR N12K showed lag time of 4 h with &gt;90% drug release. The data were statistically analyzed using ANOVA and P &lt; 0.05 was statistically significant. Release kinetics of the optimized formulation best fitted the zero order model. Optimization studies on compression coated floating-pulsatile drug delivery of bisoprolol

The physicochemical properties of spray dried micro-complexes of cefuroxime axetil (CFA) with hydroxypropyl-$\beta$-cyclodextrin (HP$\beta$CD) in presence and/or absence of ternary components polyvinylpyrrolidone K30 (PVP K30), hydroxypropylmethylcellulose (HPMC), poloxamer 188 and/or polyethylene glycol 4000 (PEG 4000) along with Aerosil{\textregistered}200 as an adsorbent were investigated. The phase solubility studies revealed AL type of solubility curve in binary as well as ternary systems. The stability constant 382.70 ± 2.4 M-1 of binary system i.e. CFA with HP$\beta$CD was significantly improved to 427.84 ± 3.8, 447.09 ± 4.3, 488.28 ± 4.6 and 502.21 ± 5.1 M-1 in presence of PVP K30, HPMC, poloxamer 188 and PEG 4000 respectively indicating positive effect of their addition. The micro-complexes of CFA were characterized by Fourier transformation infrared spectroscopy, X-ray powder diffractometry, differential scanning calorimetry, scanning electron microscopy, particle size analysis and dissolution. In all characterization studies, ternary systems performed better in comparison to binary systems as a result of synergistic effect of ternary complexation as well as particle size reduction achieved by a spray drying technology. {\textcopyright} 2012 Springer Science+Business Media B.V.

Physicochemical characterization of spray dried ternary micro-complexes of cefuroxime axetil with hydroxypropyl-β-cyclodextrin

Several methods can be approached to modify the physico-chemical properties of the drug. Here in this research work the designated supramolecularsynthon approach, with a strategy to exploit acid-amide and acid-chloride heterosynthons has been used to prepare Aceclofenac-Sodium Saccharin molecular complexes. The molecular complexes were prepared by solvent-drop grinding (ACF-SC-PVP), Mass analysis has been used to study the prepared molecular complexes. The results conclude the successful formation of the ACF- SC-PVP molecular complexes based on appearance of single DSC endothermic peak and changes observed in the vibrational frequencies associated with amide, acid and chloride groups of the reactants in FT-IR, proved to be effective diagnostic features to confirm the formation of molecular complexes. {\textcopyright} 2013 IEEE.

Cocrystal formulation, characterization and evaluation study

The aim of present study was to develop and characterize polymeric micelle (PMs) of anticancer drug, Capecitabine (CTB). Preliminary trial batches were employed to screen the significant formulation and process variables of CTB loaded polymeric micelles by Quality-by-Design (QbD) approach. Eight variables were screened and the results showed Eudragit S 100 and HP $\beta$ Cyclodextrin (HP $\beta$-CD) to be the most significant factors influencing mean particle size and zeta potential of the polymeric micelle formulation. The main effects plot depicted that all the independent variables had a statistically significant effect on the response variables. The selected batch was then studied for different characterization parameters and for invitro cytotoxicity studies. {\textcopyright} 2013 IEEE.

Capecitabine loaded polymeric micelles: Formulation, characterization and cytotoxicity study

Repaglinide is a widely used meglitinide class drug to treat diabetes. The current study describes degradation as per ICH guidelines (hydrolysis, photolysis, dry heat and oxidation) and optimized assay method. A simple stability indicating method has been developed for Repaglinide in bulk drug. The peak purity of the drug was observed. For HPTLC study Aluminium plates precoated with Silica Gel 60 F254 was the stationary phase and mobile phase consisted of methanol: toluene (2:8) and quantitation was done at the wavelength 242nm.
