The objective of this study was to design wax matrix tablets for oral sustained release of Metoprolol succinate and to investigate the sustained release behavior of the fabricated tablets. Matrices were prepared by melt granulation technique using Hydrogenated vegetable oil, Compritol and Precirol as a release retardant material. The FT-IR study indicated the compatibility of drug with excipients. The effect of different wax materials, different ratio of wax materials and excipients on release profile of metoprolol succinate was also studied. Tablet matrices containing dicalcium phosphate has given more retardant effect on release of drug than other excipients. One of the best formulations further studied for sintering time and temperature effect using direct compression method. Matrix tablet showed no change in physical appearance, drug content after storage 400C/ 75% RH for 3 months. {\textcopyright} RJPT All right reserved.

Sustained release wax matrix tablet of Metoprolol succinate

Present work describes the synthesis of (2E)-1-(8-hydroxyquinolin-7-yl)-3-(Aryl) prop-2-en-1-one, derivatives of Chalcones. Starting material 8- hydroxy quinoline was acetylated and subjected to fries rearrangement reaction to yield 7- acetaldehyde-8-hydroxy quinoline which on aldol condensation with various derivatives of benzaldehyde yields 13 compounds. The compounds have been characterized and evaluated for anti-bacterial activity. MIC values of the compounds showing higher zone of inhibition was found to be 100 $\mu$g/ml. The activity of the compounds can be attributed to the substitution of electronegative groups in the synthesized compounds. {\textcopyright} RJPT All right reserved.

Synthesis and antibacterial activity of some new (2E)-1-(8-hydroxyquinolin-7-yl)-3-(Aryl) prop-2-en-1-one, derivatives of Chalcone

Background: Renal epithelial cell injury by reactive oxygen species is pre-requisite step in the pathogenesis of urolithiasis. Rutin and curcumin are polyphenolic compounds known to have antioxidant and anti-inflammatory activities, but their effect on urolithiasis is yet to be elucidated. In the present study, we have investigated the inhibitory effect of rutin and curcumin on calcium oxalate urolithiasis in Wistar albino rats. Methods: Calcium oxalate urolithiasis was induced experimentally by administration of 0.75% v/v ethylene glycol with 1% w/v ammonium chloride in drinking water for three days followed by only 0.75% v/v ethylene glycol for 25 days. Rutin (20 mg/kg body weight) and curcumin (60 mg/kg body weight) were given once daily for 28 days by oral route. After treatment period, calcium and oxalate levels in urine and kidney tissue homogenate were measured. Kidney was also used for histopathological examination. Results: Stone-induction with ethylene glycol and ammonium chloride resulted in elevated levels of calcium and oxalate in the urine and kidney sample, whereas supplementation of rutin and curcumin restored it near to normal. Histopathological study revealed minimum tissue damage and less number of calcium oxalate deposits in kidney of animal treated with rutin and curcumin as compared to calculi-induced animal. Conclusion: The data suggest that the rutin and curcumin inhibits calcium oxalate urolithiasis. This effect is mediated possibly through a lowering of urinary concentration of stone forming constituents, anti-inflammatory and antioxidant effects.

Inhibitory effect of rutin and curcumin on experimentally-induced calcium oxalate urolithiasis in rats

Diabetes mellitus is a major global metabolic disorder of current century. Plants represent a vast source of potentially useful dietary supplements for improving blood glucose control and preventing long-term complications in type 2 diabetes mellitus. This study was undertaken to investigate the microscopic characteristics of Momordica charantia fruits and phytochemical analysis of extracts. Glucose tolerance test, hypoglycemic, antidiabetic effect, serum lipid profile and histopathological studies of pancreas, kidney and liver in normal control rats, diabetic control rats and extract treated rats were also carried out for single and repeated oral administration of the aqueous extract of Momordica charantia (Family: Cucurbitaceae) fruits in normal and alloxan induced diabetic rats. A range of doses, viz. 250 and 500 mg/kg b.w. of aqueous fruit extract of Momordica charantia were evaluated and the dose of 500 mg/kg was identified as the most effective dose. In acute studies it lowers blood glucose level around 42%after 3 h of administration and in sub-acute studies it showed 22% reduction in blood glucose level in normal rats. The same dose of 500 mg/kg produced a fall of 30% in blood glucose level within 90 min during glucose tolerance test (GTT) in normoglycemic rats. This dose has closer effect as that of standard drug glibenclamide (10 mg/kg b.w.). In diabetic rats in acute studies it lowers blood glucose level around 4%after 5 h of administration and in sub-acute studies it showed 39% reduction in fasting blood glucose level on 7th day.Total cholesterol (TC), low density lipoprotein (LDL), albumin and triglyceride (TG) levels also decreased in severely diabetic rats whereas, cardio protective, high density lipoprotein (HDL) and protein level in serum were increased. Concurrent histological studies of the pancreas of these animals showed comparable regeneration by aqueous extract and kidney and liver showed normal structure in the rats which were earlier, necrosed by alloxan. These results clearly indicate that aqueous extract of Momordica charantia has high antidiabetic potential along with significant hypoglycemic and antidiabetic effects. Further studies are warranted to fractionate the active principles and development of polyherbal formulations by using other traditional antidiabetic plant extracts and determination of its synergistic effects.

Progress of the Indian pharmaceutical industry: A shifting perspective

A review. The soly. issues present serious challenges to the successful development and commercialization of new drugs in the pharmaceutical industry. Lipophilic and less water-sol. therapeutic agents cause decreased bioavailability, increased chance of food effect, incomplete release from the dosage form and higher inter-subject variability. Self Micro-emulsifying Drug Delivery System (SMEDDS) is a novel approach to improve water soly. and ultimate bioavailability of drugs. SMEDDS is isotropic (one phase system) mixt. of oil or modified oils, surfactants and co-surfactants, which form the fine oil-in-water microemulsion when introduced into aq. phase under condition of gentle agitation. The digestive motility of the stomach and intestine provide the agitation necessary for self-microemulsion in-vivo. Triglyceride is the one of the component of SMEDDS, which helps in the absorption of drugs from the GI tract. SMEDDS enhance the bioavailability enabling redn. in dose of the drug. SMEDDS is evaluated by various methods like visual assessment, droplet polarity and droplet size, size of emulsion droplet, dissoln. test, charge of oil droplets, viscosity detn., in-vitro diffusion study. Self Micro-emulsifying Drug Delivery System: a novel approach for hydrophobic drugs.

FLOATING PULSATILE DRUG DELIVERY SYSTEM OF CAPTOPRIL BASED ON AN IMMPERMEABLE CYLINDER.

A simple, precise and accurate methods for simultaneous determination of Cefixime (CEF) and Ofloxacin (OFL) in combined tablet dosage form has been developed. The method is based on Ratio spectra derivative and Area under curve (AUC) spectrophotometry using methanol and 0.1 N HCl, respectively as solvents. The amplitudes at 319.11 nm and 347.40 nm in the first derivative of the ratio spectra were selected to determine CEF and OFL, respectively and wavelength ranges of 277-279 nm and 296-298 nm were selected to determine CEF and OFL by AUC method in combined formulation. Beer's law is obeyed in the concentration range of 5-25 μg/mL and 4-20 μg/mL by Ratio spectra derivative and Area under curve method, respectively for both the analytes. The % assay in commercial formulation was found to be in the range 99.01 - 100.90 % for CEF and 98.91 - 101.72 % for OFL by the proposed methods. The methods were validated with respect to linearity, precision and accuracy. Recovery was found in the range of 98.60 - 101.80 % for CEF and 98.75 - 100.2% for OFL by ratio derivative method and 98.16 - 100.4% for CEF and 98.89-100.21% for OFL by AUC method respectively for both the Formulations. The methods developed are simple, economical, precise and accurate and can be used for routine quality control of combined tablets. AUC method was successfully applied to carry out dissolution study of commercial tablet formulation by using USP II dissolution test apparatus.

Spectrophotometric simultaneous determination of cefixime and ofloxacin in combined tablet dosage form by ratio derivative and area under curve method and it's application to dissolution study

Two simple, rapid, accurate and economical spectrophotometric methods are described for the determination of Drotaverine (DRT) and Etoricoxib (ETR) in combined dosage form. The first method (Method A) is based on Ratio Spectra Derivative was measurement of amplitude of first order derivative of ratio spectra where DRT and ETR were determined at $\lambda$ max 268.19 nm and 338.91 nm, respectively and methanol was used as solvent. For Absorption Corrected method (Method B) in which DRT and ETR exhibit $\lambda$ max at 360.90 nm and 249.10 nm, respectively and 0.1 N HCL was used as solvent. Both the drugs obey Beer's law in the concentration ranges of 8-40 $\mu$g mL -1 for DRT and 9-45 $\mu$g mL -1 for ETR. The results of analysis have been validated statistically and recovery studies confirmed the accuracy and reproducibility of the proposed method which were carried out by following ICH guidelines. Simultaneous spectrophotometric determination of drotaverine and etoricoxib in combined tablet dosage form by ratio derivative and absorption corrected method and its application to dissolution study

Metformin hydrochloride (MET) sustained-release solid dispersions (SD) were prepared by the solvent evaporation and closed melt method, using compritol 888 ATO as the polymer with five different drug-carrier ratios. Characterization of solid dispersion was carried out by Fourier Transform Infrared (FTIR) spectroscopy, ultraviolet (UV) spectroscopy, Differential scanning calorimetry (DSC), X-ray powder diffraction (XRPD). The FTIR and UV studies suggested that no bond formation had occurred between the polymer and the drug. DSC and XPRD results ruled out any interaction or complex formation between the drug and the polymer. The formulated SD had acceptable physicochemical characters and SD with a 1: 4 drug: Polymer ratio, which released the drug over an extended period of eight-to-ten hours. The data obtained from the in vitro release studies were fitted with various kinetic models and were found to follow the Korsmeyer-Peppas equation. The prepared SD showed good stability over the studied time period. The solvent evaporation method was found to be more helpful than the closed melt method, giving the sustained release action. The SD with a 1: 4 ratio of drug to polymer, by the solvent evaporation method, was selected as the most effective candidate for the subsequent development of a well-timed, sustained-release dosage form of the drug.
