We investigate methodologies for improving the results of Physics Informed Neural Networks-Neural Networks that are trained with the supervision of relevant physical laws to solve specific problems. The methods we look into in this work are implementing the ResNet architecture and implementing Fourier Feature Mapping to allow the network to learn high-frequency features. We implement these methods to train a PINN on the 1D Burger's Equation. The predictions of the PINNs are compared against the solution from a Finite Difference Method based solver along with a baseline performance of a simple densely connected network. © 2022 IEEE.

Physics Informed Neural Networks-A Methodology Review

Diabetes mellitus is the most widely recognized endocrine disorder which is influencing a bigger populace on the planet. There are various causes of diabetes, such as physical inactivity, obesity, family history, race, and age. Diabetes mellitus is associated with some life-threatening complications, such as neuropathy, nephropathy, various eye diseases or retinopathy, and cardiovascular disorders. Many synthetic antihyperglycemic agents are available in the market for the treatment of diabetes and its complications. But, due to some serious side effects of these synthetic agents, people are opting for herbal remedies and, therefore, they are now becoming popular. Herbal remedies have lesser side effects and higher affordability and therefore can be preferably used over synthetic agents for a long-term disorder like diabetes mellitus. In the present study, scientific research and review studies on the topic were collected from Science Direct, Scopus, PubMed, Google Scholar, and other relevant sources. The references of all the articles were screened manually for any additional information on popular polyherbal formulations in traditional Ayurvedic, Chinese, and Unani medicinal systems. It is found that these polyherbal formulations are studied for anti-diabetic potential. Furthermore, some are also investigated for mechanism of action of anti-diabetic effects. This review highlights various Ayurvedic, Chinese, and Unani polyherbal formulations commonly utilized in the management of diabetes mellitus along with their pre-clinical and clinical investigations, which will enhance the existing knowledge of the researchers. Common Ayurvedic, Chinese traditional and Unani antidiabetic formulations- a review

The ascent in cancer cases is startling, with breast cancer epidemiology extending across the globe. The severity of the aggressiveness of the disease is abetted by many aspects that have a critical impact on breast cancer biology, of which hypoxia is one. There is documentation of its disparate implications that defines its far-reaching impact on the therapeutic regimen. Nanomedicine mediated therapy has been seen to alleviate the adverse impact of hypoxic tumors, which has been backed by many investigative studies. This chapter entails the elemental understanding of hypoxia, its serious fall back on disease presentation, and also acknowledges the resistance to cancer cells along with its consequential impact on the disease prognosis. In this wake, this chapter recapitulates the studies primarily emphasizing the hypoxia-mediated nanoparticle treatment scheme that has advanced to avert this grave condition that makes the cancer therapy cumbersome. © 2022 Elsevier Inc. Targeted Nanomedicine for Breast Cancer Therapy

Hypoxia mediated targeted nanomedicine for breast cancer

Octreotide acetate (OA), a potent octapeptide, is used in the treatment of pituitary adenoma. An approach has been made in the present research to formulate an OA-loaded intranasal in situ gel (OA-ISG) to target pituitary adenoma. To achieve the objective of the present work, OA-ISG was fabricated using cold method, and further optimization was done by 32 factorial design. The optimized formulation was evaluated for gelation temperature, mucoadhesive strength, and % drug release (8 h), and the results were found to be 30.01 ± 0.4 °C, 40.12 ± 0.5 g, and 98.54 ± 0.45 %, respectively. Brain availability of OA was determined through gamma scintigraphy, wherein Cmax for technetium (99mTC) labeled intranasal OA-ISG (99mTC-OA-ISG) was found to be 1.041 % RA/g, and the findings for 99mTC-OA-Solution (intranasal) and 99mTC-OA-Solution (intravenous) were 0.395 % and 0.164 % RA/g, respectively. Consequently, a 3–10-fold increase in brain OA concentrations was observed upon intranasal administration (OA-ISG) as compared to others. Additionally, drug targeting index (100.13), targeting efficiency (10013 %), and direct transport percentage (2564.1 %) corroborate brain targeting of OA via intranasal route. Further, the cytotoxic potential of OA-ISG was screened on human pituitary tumor (GH3) cell lines using MTT assay. The IC50 value was found to be 9.5 μg/mL for OA-ISG, whereas it was 20.1 μg/mL for OA-Solution, thereby confirming the superior results of OA-ISG as compared to OA-Solution. Hence, the developed intranasal OA-ISG can be further explored for establishing its potential clinical safety, and as effective platform for targeted drug delivery to the brain in pituitary adenoma. © 2022 Elsevier B.V.

Nose-to-brain delivery of octreotide acetate in situ gel for pituitary adenoma: Pharmacological and in vitro cytotoxicity studies

Triphala is a popular Ayurvedic herbal formulation. It is composed of dried fruits of Terminalia chebula, Terminalia bellirica and Phyllantus emblica. Triphala is rich in polyphenolic constituents. It is widely used in traditional systems of medicine including Ayurveda for various health benefits. Triphala is extensively studied to support its traditional claims and has been documented for antibacterial, cardioprotective, anticancer, immunomodulatory, neuropsychological, anti-inflammatory, antidiabetic, antihyperlipidemic, anti-obesity and laxative properties. These effects are mainly attributed to the presence of polyphenolic constituents in the Triphala. Hematological toxicity or pancytopenia produced by carboplatin chemotherapy tends to limit the clinical usage of carboplatin as a potent antineoplastic agent. The current investigation was aimed at evaluation of the protective activity of a plant phenolic acid i.e. 2-pyrocatechuic acid (2,3 dihyroxybenzoic acid) against hematological adverse effects induced by carboplatin in Wistar rats. Intraperitoneal injection carboplatin (35 mg/kg) for 6 days resulted in significant alterations in hematological parameters such as decreased red blood cell, hemoglobin, total leukocyte and differential leukocyte counts and increased eosinophil counts in rat blood indicating severe hematological toxicities. Pre and co-administration of 2-pyrocatechuic acid at the doses 10, 30 and 100 with CP significantly attenuated the altered hematological indices in a dose-dependent manner exhibiting its efficacy as a protective agent for the amelioration and prevention of hematological toxicities induced by carboplatin chemotherapy. © RJPT All right reserved.

2-Pyrocatechuic Acid Attenuates Carboplatin Induced Hematological Toxicities in Wistar Rats

Fenugreek has been well documented for its broad range of benefits in managing several disorders and ailments, including neurological disorders in traditional and modern scientific literature. This chapter is a comprehensive review of the scientific publications that evaluated the fenugreek against several disorders of neurological and psychological origin. The present evidence suggested fenugreek (powder, extracts, fractions, and phytoconstituents) can provide beneficial effects against neurological disorders (neurodegeneration, convulsions, centrally mediated pain, and neuropathies) and psychological disorders (stress and stress-induced disorders such as anxiety, depression, and cognitive disorders). Fenugreek: Traditional and Modern Medicinal Uses

Fenugreek in Management of Neurological and Psychological Disorders

The current study presents the synthesis and evaluation of zinc oxide and silver oxide hybrid nanoparticles for their potential apoptotic and anti-cancer activity. Owing to their small particle size, high surface area, and synergistic effects of embedded biomolecules, microwave-assisted synthesised nanoparticle-based drug delivery systems have significant potential in the field of biopharmaceutics and medicine. The higher activity of the hybrid nanoparticles may be attributed to their spherical shape, crystalline nature and lower particle size of 10–100 nm, as shown by different characterization methods as UV, FTIR, Zeta potential, particle size analysis, XRD, SEM, TEM, ICP-OES examination. The cell cycle analysis, cell death, and apoptosis in breast cancer cells (MCF-7) treated with synthesized hybrid NPs were evaluated in this work. Investigation of cell cycle analysis using flow cytometry has been utilized to analyse different phases of cell division and proliferation. The cytotoxic effect of microwave-assisted synthesised hybrid nanoparticles on MCF-7 cell line was studied using MTT assay and found to have significant cytotoxic activity with an IC50 value of 24.31 µg/ml when compared to standard drug Doxorubicin. Moreover, in vivo toxicity performed in Wistar rats reveals safety of developed hybrid nanoparticles. The findings demonstrate the powerful therapeutic efficacy of synthesized hybrid nanoparticles as well as the potential for development of future anticancer medication. © 2022 Elsevier B.V.

Evaluation of apoptotic and anti-cancer efficacy of microwave assisted (ZnO-Ag2O) hybrid nanoparticles in MCF-7 breast cancer cell lines

Over the decades the outcome of industrial compliance, manufacturing process, formulation and development, Hot Melt Extrusion (HME) also expanded the acceptance. As a result of industrial adaptability, it has developed itself into a wide range pharmaceutical research development. New drug entities face failures or delays during formulation development, including poor biopharmaceutical properties toxicity, lack of efficacy, or both. To overcome such obstacles, the HME technique is preferred one. In comparison to conventional techniques, its use is increasing because of its process, solvent free nature, cost effective, and on-line manufacturing. Recent Advances in Hot Melt Extrusion and Its Applications

The upsurging human population subsequently elevate the demand for food thereby creating immense strain on the agriculture sector worldwide. Varied types of biotic and abiotic factors are responsible for impairing the plant’s health, therefore efficacious management strategies for these factors are required for food security. The diverse conventional methods for controlling plant diseases come up with numerous unavoidable limitations. The development of novel competent strategies which can provide the solutions for plant disease management needs more attention. Herein, we expanses the knowledge about the new field that is nanophytovirology to specifically combat plant viral diseases. Nanophytovirology is the budding area attracting more attention day by day because the area can control plant viral diseases with efficient well-versed methods that involve the use of nano-sized materials. Nanomaterials can be used as antivirals, as delivery systems, as biostimulants and also as sensitive and selective detection systems for phytoviral infections. Mainly various metallic and metal oxides nanoparticles were explored for their antiviral activity, although nanomaterial delivery systems for combating viral diseases need to be considered more. However, the potentials of nanomaterials as delivery systems are very well highlighted in the chapter. Further, nanomaterials have a great ability to enhance plant immunity by their biostimulation phenomenon. The nanomaterials are ideal agents to effectively diagnose and detect plant viral diseases with the help of functional, portable, rapid, sensitive and selective nanobiosensors. © 2023 Sunil K.
